摘　　要：目的通过生物信息学方法研究肝细胞癌(Hepatocellular carcinoma,HCC)中铜死亡相关lncRNA与肿瘤预后的联系。方法从TCGA数据库下载HCC样本转录组、基因突变和相应的临床数据,通过共表达分析获得铜死亡相关lncRNA,采用Cox回归分析和LASSO分析构建HCC患者预后模型并对其预测能力进行评价。结果构建包含7个铜死亡相关lncRNA(AC105020.1、CHASERR、FOXD2-AS1、MIR4435-2HG、NRAV、RNF216P1、SNHG3)的预后模型,Kaplan-Meier生存曲线和ROC曲线表明该模型具有准确的预测能力。Cox回归分析显示模型风险评分为影响HCC患者预后的独立危险因素(P<0.05)。肿瘤突变负荷在高、低风险组间差异有统计学意义,高风险组HCC患者对免疫治疗的反应优于低风险组。结论本研究构建的铜死亡相关lncRNA预后模型能够较好地预测患者预后,为HCC患者个体化治疗提供参考。Objective To study the relationship between cuprotosis-related lncRNAs and tumor prognosis in hepatocellular carcinoma(HCC)by bioinformatics.Methods The transcriptome,genomic mutations and corresponding clinical data of HCC patients were downloaded from the TCGA database,and the cuprotosis-related lncRNAs was obtained by co-expression analysis.Cox regression and LASSO regression were used to construct a prognosis model of HCC patients and evaluate its predictive ability.Results The seven cuprotosis-related lncRNAs prognostic models were constructed(AC105020.1,CHASERR,FOXD2-AS1,MIR4435-2HG,NRAV,RNF216P1,SNHG3).The Kaplan Meier survival curve and ROC curve showed that the model had accurate predictive ability.Cox regression analysis showed that risk score was an independent risk factor affecting the prognosis of HCC patients(P<0.05).There were significant differences in tumor mutation burden between the high-and low-risk groups.HCC patients in the high-risk group responded better to immunotherapy than those in the low-risk group.Conclusion The cuprotosis-related lncRNAs prognostic model constructed in this study can better predict the prognosis of HCC patients,which may be a reference for the individualized treatment of HCC.
