摘  要:以鲁氏酵母菌QOR(Quinone oxidoreductase)过表达工程菌为对象,探讨了对苯二酚和1,4-萘醌对酵母菌合成呋喃酮产量、QOR基因表达量及菌体的影响。结果表明,对苯二酚和1,4-萘醌的添加浓度分别为10μg·mL~(-1)和10 mg·L^(-1)、培养时间为3和5 d时,酵母菌的呋喃酮产量均显著高于对照组(P<0.05)。两种外源添加物显著提高了酵母菌QOR基因相对表达量(P<0.05),添加1,4-萘醌的酵母菌细胞数高于对苯二酚组,对酵母菌的细胞形态影响较小。因此,添加对苯二酚和1,4-萘醌有助于酵母菌的生长,可显著促进鲁氏酵母工程菌代谢合成呋喃酮。In order to study the effects of hydroquinone and 1,4-naphthoquinone on the quinone oxidoreductase(QOR)gene overexpression engineering strain Zygosaccharomyces rouxii and the content of furanone production,QOR gene expression and the changes of cell morphology and quantity were determined.The results showed that when hydroquinone and 1,4-naphthoquinone were added at 10μg·mL-1 and 10 mg·L^(-1) and cultured at 3 d and 5 d,respectively,the yields of furanone were significantly higher than those of the control(P<0.05).Two exogenous additives could significantly increase the relative expression level of QOR gene(P<0.05)and did not affect the cell morphology of engineered strain Z.rouxii.The number of yeast cells with 1,4-naphthoquinone addition was slightly higher than that of hydroquinone.Therefore the addition of hydroquinone and 1,4-naphthoquinone could facilitate the growth of yeast and significantly promote the metabolism and synthesis of furanone by engineered strain Z.rouxii.
