外泌体非编码RNA在腹主动脉瘤患者中的表达及其临床意义

摘  要:目的 筛选出正常健康人和腹主动脉瘤患者外泌体中表达有差异的非编码RNA(ncRNA),并进行生物信息学分析以了解其内部机制。方法 收集健康对照组和腹主动脉瘤患者的血浆,提取外泌体中的ncRNA并进行高通量测序。表达差异采用DEseq进行评估,并根据|log2(折叠变化)|>1和调整后的P<0.05的标准进行筛选并排序。基于《京都基因与基因组百科全书》(KEGG)和生物通路与基因本体论(GO)进行功能富集分析。结果 筛选了621种差异表达的ncRNA,显著上调的前10个ncRNA包括MIR1539、MIR3674、SNORD115-47、MIR3188、MIR4443、microRNA 7641-1、MIR4262、SNORD115-18、MIR5096、MIR557;显著下调的前10个ncRNA包括SCARNA13、SNORA11、SNORD3A、RNU4-1、RMRP、RNU4-2、MALAT1、NR144567.1、TALAM1、SNORA73B。发现了与这些基因显著相关的通路,即内质网中的代谢通路、钙信号通路和蛋白加工通路。结论 正常人与腹主动脉瘤患者中外泌体ncRNA的表达有差异,外泌体可能通过激活ncRNA代谢途径和钙信号通路调控腹主动脉瘤的发生与进展。外泌体中某些差异表达的ncRNA可能作为腹主动脉瘤的新型生物标志物。Objective Non-coding RNAs(ncRNAs)with statistically different expression in exosomes from normal healthy subjects and patients with abdominal aortic aneurysms were screened and bioinformatically analyzed to understand their internal mechanisms.Methods Plasma was collected from healthy controls and patients with abdominal aortic aneurysms,and non-coding RNAs in exosomes were extracted and subjected to high-throughput sequencing.Expression differences were assessed using DEseq and screened and ranked according to the criteria of|log 2(fold change)|>1 and adjusted P<0.05.Functional enrichment analysis was performed based on the Kyoto Encyclopedia of Genes and Genomes(KEGG)and biological pathways and gene ontology(GO).Results A total of 621 differentially expressed ncRNAs was screened,and the top 10 significantly up-regulated ncRNAs included MIR1539,MIR3674,SNORD115-47,MIR3188,MIR4443,microRNA 7641-1,MIR4262,SNORD115-18,MIR5096,and the top 10 ncRNAs significantly down-regulated included SCARNA13,SNORA11,SNORD3A,RNU4-1,RMRP,RNU4-2,MALAT1,NR144567.1,TALAM1,SNORA73B.Pathways significantly associated with these genes were identified,namely metabolic pathways in the endoplasmic reticulum,calcium signaling pathways,and protein processing pathways.Conclusion There are significant differences in the expression of exosomal non-coding RNAs between normal subjects and patients with abdominal aortic aneurysms,and exosomes may regulate the development and progression of abdominal aortic aneurysms through activation of non-coding RNA metabolic pathways and calcium signaling pathways.Some differentially expressed non-coding RNAs in exosomes may serve as novel biomarkers of abdominal aortic aneurysms.
