摘  要:目的研究鼻咽癌(NPC)组织中微小RNA(miR)-107、叉头盒蛋白M1(FoxM1)的表达,分析二者表达与上皮间质转化(EMT)基因表达的相关性及与患者临床病理特征的关系。方法选取79例NPC患者,RT-qPCR法检测NPC组织及癌旁组织中miR-107、FoxM1 mRNA、EMT通路基因N钙黏素(N-cad)、E钙黏素(E-cad)及锌指转录因子(Snail)mRNA表达。采用t检验分析两者表达与患者临床病理特征的关系,Pearson线性相关分析miR-107与FoxM1 mRNA表达的相关性及miR-107、FoxM1 mRNA表达与EMT通路基因表达的相关性。结果NPC组织中FoxM1、N-cad、Snail mRNA的相对表达量均高于癌旁组织,而miR-107、E-cad mRNA的相对表达量均低于癌旁组织(P均<0.05)。NPC组织中miR-107、FoxM1表达与TNM分期有关(P均<0.05),而与性别、年龄、吸烟史、组织分化程度及是否伴有淋巴结转移无关(P均>0.05)。miR-107和FoxM1的表达呈负相关关系(r=-0.603,P<0.05)。NPC组织中miR-107表达与E-cad mRNA表达呈正相关关系(r=0.627,P<0.05),与N-cad、Snail mRNA表达均呈负相关关系(r分别为-0.724、-0.621,P均<0.05)。FoxM1 mRNA表达与E-cad mRNA表达呈负相关关系(r=-0.537,P<0.05),与N-cad、Snail mRNA表达均呈正相关关系(r分别为0.634、0.736,P均<0.05)。结论NPC组织中miR-107表达降低,而FoxM1表达增高,miR-107及FoxM1可能通过促进EMT通路基因表达,促进NPC进展,有可能成为新的NPC肿瘤标志物。Objective To study the expression of microRNA(miR)-107 and forkhead box M1(FoxM1)in nasopha⁃ryngeal carcinoma(NPC),and to analyze their relationships with the expression of epithelial-mesenchymal transition(EMT)pathway gene and the clinicopathological characteristics of NPC patients.Methods Seventy-nine NPC patients were selected.The expression of miR-107,FoxM1 mRNA and EMT pathway genes N-cadherin(N-cad),E-cadherin(E-cad),and Snail mRNA in NPC tissues and adjacent tissues were detected by real-time quantitative PCR(RT-qPCR).The relationship between the expression of the two and clinicopathological characteristics was statistically analyzed by t-test.Pearson linear correlation analysis was used to analyze the correlation between miR-107 and FoxM1 mRNA expres⁃sion and the correlation between miR-107,FoxM1 mRNA expression and EMT pathway gene expression.Results The relative expression of FoxM1,N-cad,and Snail mRNA in NPC cancer tissues was higher than that in the adjacent tissues,while the relative expression of miR-107 and E-cad mRNA was lower than that in the adjacent tissues,with statistically sig⁃nificant differences(all P<0.05).The expression levels of miR-107 and FoxM1 in NPC tissues were related to the tumor TNM stage(both P<0.05),but were not related to gender,age,smoking history,tumor differentiation,or whether there was lymph node metastasis(all P>0.05).The expression of miR-107 and FoxM1 showed a significant negative correlation(r=-0.603,P<0.05).In NPC cancer tissues,miR-107 was significantly positively correlated with E-cad mRNA(r=0.627,P<0.05),and was negatively correlated with N-cad mRNA and Snail mRNA(r=-0.724,-0.621,both P<0.05).FoxM1 mRNA was negatively correlated with E-cad mRNA(r=-0.537,P<0.05),and was positively correlated with N-cad mRNA and Snail mRNA(r=0.634,0.736,both P<0.05).Conclusion The expression of miR-107 in NPC cancer tissue decreases,while the expression of FoxM1 increases;miR-107 and FoxM1 may promote the tumor progression of NPC by promoting the expression of EMT pathway gen
