摘  要:目的 优化硒化大枣多糖的制备工艺,并研究其抗氧化和抗疲劳作用。方法 采用硝酸-亚硒酸钠法修饰制备硒化大枣多糖,以内黄大枣多糖为原料,以硒含量为指标,通过单因素实验和Design-Expert软件设计响应面实验确定硒化大枣多糖的最佳制备工艺。同时研究其对1,1-二苯基-2-苦基肼(1,1-diphenyl-2-bitter hydrazine, DPPH)自由基和羟基自由基清除能力和对小鼠的抗疲劳作用。结果 最佳制备工艺条件为:硝酸浓度0.5%、反应温度70℃、反应时间8.2 h,该条件下,硒含量为2.53%。体外抗氧化实验表明,当样品质量浓度为5 mg/mL时,硒化大枣多糖对DPPH自由基和羟基自由基清除率分别为83.36%和79.29%,均高于大枣多糖;硒化大枣多糖对DPPH自由基和羟基自由基的半抑制浓度(half maximal inhibitory concentration, IC50)值分别为0.799和1.145,表明硒化大枣多糖具有较强的抗氧化能力。小鼠的抗疲劳实验表明,硒化大枣多糖能够增加小鼠游泳时间,降低小鼠体内的血乳酸(blood lactic acid, BLA)和血尿素氮(blood urea nitrogen, BUN)含量,增加小鼠体内的肝糖原(hepatic glycogen, HG)和肌糖原(muscle glycogen, MG)含量。结论 该研究获得了硒化大枣多糖的最佳制备工艺,得到的硒化大枣多糖具有较强的抗氧化和抗疲劳作用。Objective To optimize the preparation process of selenified Ziziphus jujuba polysaccharides and study its anti-oxidation and anti-fatigue activity.Methods The selenized Ziziphus jujuba polysaccharides were prepared using the nitric acid sodium selenite method,using Neihuang Ziziphus jujuba polysaccharides as raw materials and selenium content as indicators.The optimal preparation process for selenized Ziziphus jujuba polysaccharides was determined through single factor experiments and Design-Expert software design response surface methodology experiments.At the same time,the scavenging ability to 1,1-diphenyl-2-bitter hydrazine(DPPH)radical and hydroxyl radical and its anti-fatigue effect on mice were studied.Results The optimum preparation process conditions were:Nitric acid concentration 0.5%,reaction temperature 70℃,reaction time 8.2 hours,under this condition,the selenium content was 2.53%.In vitro anti-oxidant test showed that when the mass concentration of the sample was 5 mg/mL,the DPPH radical and hydroxyl radical scavenging rates of selenized Ziziphus jujuba polysaccharides were 83.36%and 79.29%,respectively,which were higher than those of Ziziphus jujuba polysaccharides;the half maximum inhibitory concentration(IC50)values of selenized Ziziphus jujuba polysaccharides on DPPH free radicals and hydroxyl radical were 0.799 and 1.145,respectively,indicating that selenized Ziziphus jujuba polysaccharides had strong antioxidant capacity.Anti-fatigue experiments in mice had showed that selenized Ziziphus jujuba polysaccharides could increase swimming time,reduce blood lactic acid(BLA)and blood urea nitrogen(BUN)content in mice,and increase hepatic glycogen(HG)and muscle glycogen(MG)content in mice.Conclusion This study obtaines the optimal preparation process for selenized Ziziphus jujuba polysaccharides,which has strong anti-oxidant and anti-fatigue effects.
