超声结合低浓度氯处理对即食蓝莓微生物多样性的干预作用

摘  要:低频超声(28 kHz)结合低浓度氯(10 mg/L)能有效灭活蓝莓表面细菌、霉菌和酵母。为探究其作用机制,本研究在第0天和第5天采用16S rRNA和ITS测序技术分析超声结合低浓度氯处理后蓝莓表面细菌和真菌多样性变化。结果表明蓝莓表面优势细菌为罗尔斯顿尼亚菌属、鞘氨醇单胞菌属、乳杆菌属,杀菌未改变三种微生物相对丰度;贮藏5 d后三种微生物仍为优势菌。优势真菌为曲霉属、赤霉属、圆孢霉属、木耳属、毕赤属,杀菌处理后为木耳属、曲霉属、毕赤属、嗜热链球菌属、马拉色菌属;贮藏5 d后,优势菌为嗜热链球菌属、木耳属、哈萨克斯坦菌属、毕赤属、曲霉属,其中嗜热链球菌属变化最为显著,与第0天相比相对丰度升高9.88%。上述结果表明超声结合低浓度氯处理可能对三种细菌灭杀效果一致,使其在0~5 d内相对丰度未发生明显变化;对真菌中曲霉属灭杀效果较好,使其相对丰度下降,而对木耳属灭杀效果相对较差,使其相对丰度升高,同时使嗜热链球菌属成为优势菌属,后续贮藏过程中该菌可能抑制了其它真菌生长。The naturally present microbes on blueberry could be effectively inactivated by the combination disinfection of low frequency ultrasound(28 kHz)and low concentration chlorine(10 mg/L),This study aim to explore the effect of combination disinfection on bacterial and fungi diversity from day 0-5,as analyzing by 16S rRNA and ITS sequencing technologies.The results showed that the dominated bacteria on blueberry were Ralstonia,Sphingosa and Lactobacillus,and the relative abundance(RA)of these three were not changed after disinfection and storage for 5 d.The dominated fungi were Aspergillus,Gibberella,Staphylotrichum,Auricularia and Pichia,whereas the dominated fungi were changed to Auricularia,Aspergillus,Pichia,Mycothermus,and Malassezia after disinfection.At 5 d,the dominated fungi were Mycothermus,Auricularia,Kazakistanella,Pichia and Aspergillus and the RA of Mycothermus was increased by 9.88%as compared with the RA at day 0.These results indicated that the disinfection efficacy of combination treatment against the three bacterial might be consistent between each other.The disinfection efficacy against Aspergillus might be better than Auricularia and thus lead the RA increase of Mycothermus to control other fungi growth during storage.
