摘  要:目的探讨决明子多糖通过调节白介素-6(IL-6)/Janus激酶2(JAK2)/信号转导与转录激活因子3(STAT3)通路对高糖诱导的视网膜神经节细胞损伤的影响。方法将视网膜神经节细胞RGC-5分为:对照组(正常培养的RGC-5细胞)、高糖组(50 mmol/L葡萄糖处理细胞)、决明子多糖低剂量组(50 mmol/L葡萄糖+15 mg/L决明子多糖)、决明子多糖高剂量组(50 mmol/L葡萄糖+60 mg/L决明子多糖)、激活剂组(50 mmol/L葡萄糖+60 mg/L决明子多糖+0.5μmol/L JAK2/STAT3激活剂colivelin)、激活剂对照组(50 mmol/L葡萄糖+0.5μmol/L JAK2/STAT3的激活剂colivelin);CCK-8法检测细胞增殖;流式细胞仪检测细胞凋亡;ELLSA法检测细胞上清液中丙二醛(MDA)水平、超氧化物歧化酶(SOD)活性和谷氨酸(Glu)含量;Western blot检测细胞中IL-6、p-JAK2/JAK2、p-STAT3/STAT3、增殖细胞核抗原(PCNA)、半胱氨酸蛋白酶3(caspase-3)、自噬微管相关蛋白轻链3(LC3)Ⅱ/LC3Ⅰ、Beclin-1的表达。结果与对照组比较,高糖组RGC-5细胞的OD450值、SOD活性、PCNA表达降低,细胞凋亡率、MDA水平、Glu含量、IL-6、p-JAK2/JAK2、p-STAT3/STAT3、caspase-3、LC3Ⅱ/LC3Ⅰ、Beclin-1表达上升(P<0.05);与高糖组比较,决明子多糖低剂量组、决明子多糖高剂量组RGC-5细胞的OD450值、SOD活性、PCNA表达升高,细胞凋亡率、MDA水平、Glu含量、IL-6、p-JAK2/JAK2、p-STAT3/STAT3、caspase-3、LC3Ⅱ/LC3Ⅰ、Beclin-1表达降低(P<0.05);加入JAK2/STAT3的激活剂colivelin后,减弱了决明子多糖对RGC-5细胞增殖的促进作用,且细胞凋亡能力、氧化应激和自噬反应增强。结论决明子多糖可通过抑制IL-6/JAK2/STAT3通路对高糖诱导的视网膜神经节细胞起到保护作用。Objective To investigate the influence of cassia polysaccharide on high glucose-induced retinal ganglion cell injury by regulating the interleukin-6(IL-6)/Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)pathway.Methods RGC-5 The retinal ganglion cell line RGC-5 was treated with blank control(control group),50mmol/L glucose(high glucose group),50mmol/L glucose+15mg/L cassia polysaccharide(low-dose cassia polysaccharide group),50mmol/L glucose+60mg/L cassia polysaccharide(high-dose cassia polysaccharide group),50mmol/L glucose+60mg/L cassia polysaccharide+0.5μmol/L JAK2/STAT3 activator colivelin(activator group)and 50mmol/L glucose+0.5μmol/L JAK2/STAT3 activator colivelin(activator control group).Cell proliferation and apoptosis were detected by CCK-8 assay and flow cytometry,respectively.The contents of malondialdehyde(MDA),superoxide dismutase(SOD)and glutamate(Glu)in cell supernatant were detected by Enzyme-linked immunosorbent assay(ELISA).Western blot was performed to measure the expression levels of IL-6,p-JAK2/JAK2,p-STAT3/STAT3,proliferating cell nuclear antigen(PCNA),cysteine protease 3(caspase-3),autophagy microtubule-associated protein light chain 3(LC3)Ⅱ/LC3I,and Beclin-1.Results Compared with those of control group,the OD450 value,SOD content and PCNA expression in RGC-5 cells of high glucose group were significantly reduced,while the apoptotic rate,contents of MDA and Glu,IL-6 level,and expression levels of p-JAK2/JAK2,p-STAT3/STAT3,caspase-3,LC3Ⅱ/LC3I,and Beclin-1 expressions were significantly enhanced(P<0.05).Compared with those of high glucose group,the OD450 value and SOD content of RGC-5 cells in low-,and high-dose cassia polysaccharide group were significantly enhanced,while the apoptotic rate,contents of MDA and Glu,IL-6 level,and expression levels of p-JAK2/JAK2,p-STAT3/STAT3,caspase-3,LC3Ⅱ/LC3I,and Beclin-1 were significantly reduced(P<0.05).The treatment of the JAK2/STAT3 activator colivelin weakened the role of cassia polysaccharides in promoting the p
