摘 要:目的探讨E-钙粘蛋白(E-cadherin)基因启动子区甲基化状态及其mRNA表达在卵巢癌发生发展中的作用。方法甲基化特异性聚合酶链反应(MSP)检测E-cadherin基因启动子区在48例卵巢癌组织及48例正常卵巢组织中的甲基化状态,荧光实时定量PCR(qRT-PCR)检测E-cadherin基因mRNA的表达情况。比较卵巢癌组织和正常卵巢组织中E-cadherin基因启动子区甲基化水平及其mRNA表达的差异情况,并分析它们与卵巢癌患者临床病理特征之间的关系。结果卵巢癌组织中E-cadherin基因启动子区甲基化阳性率显著高于正常卵巢组织(P<0.05),E-cadherin基因mRNA表达水平显著低于正常卵巢组织(P<0.05),且E-cadherin基因启动子区甲基化阳性的卵巢癌组织中E-cadherin mRNA表达量显著低于甲基化阴性的卵巢癌组织(P<0.05)。此外,卵巢癌组织中E-cadherin基因启动子区甲基化状态及其mRNA表达水平与卵巢癌患者的FIGO分期及淋巴结转移显著相关(P<0.05)。结论E-cadherin基因启动子区的高甲基化导致其表达下调可能在卵巢癌发生发展中起重要作用,E-cadherin基因甲基化状态有可能成为卵巢癌分子诊断和治疗的重要靶点。Objective To analyze the function of methylation status and mRNA expression of E-cadherin in the pathogenesis of epithelial ovarian cancer.Methods The methylation status of E-cadherin gene promoter was investigated in 48 ovarian cancer tissues and 48 normal ovarian tissues using methylation-specific polymerase chain reaction(MSP).The mRNA expression of E-cadherin was evaluated using quantitative reverse transcription polymerase chain reaction(qRT-PCR).The methylation profile and mRNA levels of E-cadherin were compared between ovarian cancer tissues and controls,and their correlations with the clinicopathological characteristics of patients with ovarian cancer were analyzed.Results The methylation levels of E-cadherin promoter region in ovarian cancer tissues was significantly up-regulated compared with that in normal ovarian tissues(P<0.05),and the mRNA expression of E-cadherin was significantly down-regulated in ovarian cancer tissues compared with that in the normal ovarian tissues(P<0.05).Moreover,the mRNA expression of E-cadherin in ovarian cancer tissues with methylation positive was significantly lower than that in the methylation negative group(P<0.05).In addition,the methylation status and expression levels of E-cadherin were significantly correlated with the FIGO stage and lymph node metastases of ovarian cancer(P<0.05).Conclusion Down-regulated mRNA level of E-cadherin due to hypermethylation of its promoter region may be involved in the development of ovarian cancer.The methylation status of E-cadherin promoter may be used as a biomarker for early detection and treatment of ovarian cancer.
