MAGE-As蛋白及mRNA在肺癌患者的表达水平变化及其临床意义

摘　　要：目的:探讨黑色素瘤相关抗原(MAGE)-As蛋白及mRNA在非小细胞肺癌中的表达及临床意义。方法:选取2015年1月至2020年1月在我院治疗的非小细胞肺癌患者88例,同时选取癌旁组织作为对照,采用Western blot检测肺癌和癌旁组织MAGE-As蛋白表达,RT-PCR检测肺癌和癌旁组织MAGE-As mRNA表达。结果:非小细胞肺癌组织MAGE-As蛋白及mRNA相对表达量分别为(0.343±0.101)和(0.728±0.112),明显高于癌旁组织(P<0.05);临床分期Ⅲ~Ⅳ期肺癌组织MAGE-As蛋白相对表达量为(0.385±0.102),明显高于Ⅰ~Ⅱ期肺癌组织(P<0.05);不同性别、年龄、肿瘤直径、病理类型、分化程度肺癌组织MAGE-As蛋白相对表达量比较差异无统计学意义(P>0.05);临床分期Ⅲ~Ⅳ期、中低分化程度肺癌组织MAGE-As mRNA相对表达量为(0.762±0.112)和(0.742±0.107),明显高于Ⅰ~Ⅱ期和高分化肺癌组织(P<0.05);不同性别、年龄、肿瘤直径、病理类型肺癌组织MAGE-As mRNA相对表达量比较差异无统计学意义(P>0.05);肺癌组织MAGE-As mRNA高表达和低表达患者中位总生存期为33个月和44个月,MAGE-As mRNA高表达和低表达患者生存率有所差异(P<0.05)。MAGE-As蛋白高表达和低表达患者中位总生存期差异比较无统计学意义(P>0.05)。结论:MAGE-As蛋白及mRNA在非小细胞肺癌中表达上调,与病情严重程度有关,同时MAGE-As mRNA高表达患者预后较差。Objective:To investigate the expression and clinical significance of melanoma associated antigen(MAGE)-As protein and mRNA in non-small cell lung cancer.Methods:A total of 88 patients with non-small cell lung cancer treated in our hospital from January 2015 to January 2020 were selected,and the adjacent tissues were selected as the control,the expression of MAGE-As protein in lung cancer and adjacent tissues was detected by Western blot,and the expression of MAGE-As mRNA in lung cancer and adjacent tissues was detected by RT-PCR.Results:The relative expression levels of MAGE-As protein and mRNA in NSCLC tissues were(0.343±0.101)and(0.728±0.112),which were significantly higher than those in adjacent tissues(P<0.05);the relative expression of MAGE-As protein in stageⅢ~Ⅳlung cancer was(0.385±0.102),which was significantly higher than that in stageⅠ~Ⅱlung cancer(P<0.05);there were no significant difference in the relative expression of MAGE-As protein in different gender,age,tumor diameter,pathological type and differentiation degree(P>0.05);the relative expression of MAGE-As mRNA in clinical stageⅢ~Ⅳand moderately and moderately differentiated lung cancer tissues were(0.762±0.112)and(0.742±0.107),which were significantly higher than those in stageⅠ~Ⅱand well differentiated lung cancer tissues(P<0.05);there were no significant difference in the relative expression of MAGE-As mRNA in different gender,age,tumor diameter and pathological type(P>0.05).The median overall survival of patients with high and low MAGE-As mRNA expression in lung cancer tissues was 33 and 44 months,with a difference in survival between patients with high and low MAGE-As mRNA expression(P<0.05).The difference in median overall survival between patients with high and low MAGE-As protein expression was not statistically significant(P>0.05).Conclusion:The expression of MAGE as protein and mRNA are up-regulated in non-small cell lung cancer,is related to the severity of the disease,and the prognosis of patients with high express
