摘 要:本文以K326(B2F)醇化烟叶为试验材料,对其进行附生菌筛选鉴定,并分析附生菌对烟叶发酵效果的影响。结果表明:采用平板分离技术分离培养出46株活体纯培养物,依次编号为CHJ1~CHJ46,纯化菌株通过蛋白酶筛选鉴别培养基和淀粉酶筛选鉴别培养基筛选培养,鉴别筛选出CHJ3、CHJ9菌株具有淀粉酶水解活性,CHJ5、CHJ28、CHJ36菌株具有蛋白酶水解活性;在上述5种菌株中,CHJ5菌株促进烟叶的发酵效果最好,发酵后的烟叶化学成分更加合理,评吸得分达到91.0分,与其他处理差异达显著水平;CHJ5菌株隶属微杆菌属(Microbacterium),暂命名为Microbacterium sp. CHJ5,可作为烟叶发酵微生物制剂开发的备用菌株,应用前景广阔。This paper took K326(B2F)aging tobacco leaves as the experimental material to screen and identify epiphyte,and analyzed the impact of epiphyte on the fermentation effect of tobacco leaves.The results showed that 46 live pure cultures were isolated and cultured using plate separation technology,numbered CHJ1~CHJ46 in sequence.The purified strains were screened and cultured using protease screening and identification medium and amylase scree-ning and identification medium.It was found that CHJ3 and CHJ9 strains had amylase hydrolysis activity,while CHJ5,CHJ28 and CHJ36 strains had protease hydrolysis activity;among the five strains mentioned above,CHJ5 strain had the best effect in promoting the fermentation of tobacco leaves.The chemical composition of the fermented tobacco leaves was more reasonable,with a smoking evaluation score of 91.0,which was significantly different from other treatments;CHJ5 strain belonged to Microbacterium,and was temporarily named Microbacterium sp.CHJ5.It could be used as a backup strain for the development of tobacco leaf fermentation microbial preparations and had broad application prospects.
