摘 要:采用闪式提取法提取麦冬皂苷。以麦冬为原料,在单因素试验的基础上,以麦冬皂苷得率为响应值,以液料比、乙醇浓度、提取时间和提取电压为考察因素,采用Box-Behnken响应面法对麦冬皂苷的闪式提取工艺进行优化,并以对DPPH自由基、ABTS+自由基、OH自由基的清除率为指标评价麦冬皂苷抗氧化活性。结果表明,闪式提取法提取麦冬皂苷的最佳工艺条件为液料比25∶1 mL/g、提取时间80s、提取电压111V、乙醇浓度71%,在此工艺条件下,麦冬皂苷的提取率为5.92 mg/g;麦冬皂苷对3种自由基均具有显著的清除作用。闪式提取法具有提取时间短、操作简便、能用于麦冬皂苷的高效提取。Ophiopogon saponins were extracted by flash extraction method.Taking Ophiopogon japonicus as raw material,based on single factor experiment,taking the yield of Ophiopogon japonicus as the response value,and the liquid-to-solid ratio,ethanol concentration,extraction time and extraction voltage as the investigation factors,the Box-Behnken response surface method was used to analyze the wheat worms.The flash extraction process of Ophiopogon saponins was optimized,and the antioxidant activity of Ophiopogon saponins was evaluated by the scavenging rate of DPPH free radicals,ABTS+free radicals and OH free radicals.The results showed that the optimal process conditions for the extraction of Ophiopogon saponins by flash extraction were as follows:the ratio of liquid to material was 25∶1 mL/g,the extraction time was 80 s,the extraction voltage was 111 V,and the ethanol concentration was 71%,under this process condition,the extraction rate of Ophiopogon saponins was 5.92 mg/g;Ophiopogon saponins had significant scavenging effects on the three free radicals;The flash extraction method has the advantages of short extraction time,simple operation,and can be used for efficient extraction of Ophiopogon saponins.
