摘  要:从江西东乡野生稻自然保护区野生稻(Oryza sativa L.)植株中分离获得1株固氮菌株Y3,通过16S rRNA和基因组系统发育分析,鉴定为Kosakonia sacchari。生理生化和促生试验表明,Y3对重金属Cu^(2+)、Ni^(2+)、Co^(2+)和Zn^(2+)具有较好的耐受性,Cd^(2+)的耐受浓度达到200μg/mL,同时具有固氮和产铁载体的功能。基因组注释和比较基因组学分析显示,Y3基因组大小为5.1 Mb,GC含量53.68%,具有5140个编码基因,其中包含74个tRNA基因、8个rRNA基因和4985个功能蛋白基因;同源基因分析表明,6株Kosakonia sacchari菌株共有3987个基因簇,其中Y3与其他5株菌株共享4417个基因簇;比较基因组分析表明,Y3基因组中存在与植物促生作用有关的铁载体合成酶和固氮酶相关基因,以及与Zn^(2+)转运和Cu^(2+)抗性相关的基因。本试验深入研究了Y3的促生和重金属耐受性机制,为更有效地利用其在植物-微生物联合修复重金属污染土壤方面提供了理论依据。A metal-tolerant strain Y3 was isolated from wild rice(Oryza sativa L.)plants in Dongxiang District of Jiangxi Province and identified as Kosakonia sacchari based on the 16S rRNA gene and genome sequence.Strain Y3 exhibited increased tolerance to heavy metals including Cu^(2+),Ni^(2+),Co^(2+)and Zn^(2+).The bacteria also showed high siderophore-producing and nitrogen-fixing capacity.Based on these important features,the whole genome was sequenced and annotated and a comparative genome analysis was conducted.The total size of the Y3 genome is 5.1 Mb,with a 53.68%GC content and 5140 coding genes,which contained 4985 predicted protein-coding genes,74 tRNA,and 8 rRNA genes.In the Venn diagram,the six Kosakonia sacchari strains had 3987 clusters and Y3 shared 4417 clusters with the other five strains.Moreover,genome annotation using the PATRICK database identified several genes that encode nitrogenase and siderophore-producing enzymes involved in plant growth promotion,the presence of genes related to Zn^(2+)transportation and Cu^(2+)resistance may explain the heavy metals tolerance of strain Y3.The wholegenome analysis of this strain clarified the genetic basis underlying its phenotypic and biochemical characteristics,underpinning the beneficial interaction between Y3 and host plants,and providing a basis for more effective use of it in the phytoremediation of heavy metal-contaminated soil.
