摘  要:DNA的完整性和稳定性对于机体的正常运转和生存至关重要,但DNA易受到诸多外源性和内源性因素的影响而造成不同程度的损伤[1]。当DNA发生损伤时,细胞内的特殊分子对损伤的识别将激活DNA损伤应答(DNA damage response,DDR)系统,通过复杂的信号传导网络募集多种修复因子到损伤部位并启动DNA损伤的修复。DNA damage response(DDR)is essential for the survival of organisms,and the regulation of DDR has become a key strategy for the treatment of many diseases,especially for cancer.As an important mediator of intercellular communication,exosomes can transport their contents into recipient cells to elicit various functions,and their application in the treatment of human diseases has been investigated extensively.In recent years,the relationship between exosomes and DDR has been gradually recognized.On one hand,DNA damage can influence the secretion and the composition of exosomes;on the other hand,exosomes up-taken by recipient cells can modulate cellular DDR,thus exhibiting a reciprocal regulation between exosomes and DDR.This review summarized the current research progress on the relationship between exosomes and DDR,the underlying mechanisms,and clinical applications for such relationship.
