摘 要:为进一步研究化梁王茶多糖,对其进行纯化后评价其体外抗氧化活性。采用Sevag法、AB-8大孔树脂、DEAE-52纤维素、凝胶分离技术对梁王茶粗多糖进行纯化;测定纯化前后多糖中多糖、糖醛酸、蛋白质、总酚和总黄酮含量变化;运用傅里叶变换红外光谱(fourier transform infrared spectroscopy,FT-IR)和扫描电镜(scan-ning electron microscope,SEM)表征其结构特点;再通过DPPH自由基清除实验和FRAP法评估其抗氧化能力。结果表明,通过纯化梁王茶多糖中的蛋白质、总酚、总黄酮被大量除去,呈现清晰的中空球体状结构,并表现了一定的抗氧化活性。纯化后多糖的DPPH自由基清除能力增强,IC50值由7.274 mg·mL^(-1)降低为7.101 mg·mL^(-1);而FRAP抗氧化能力则略有降低,抗氧化能力由0.342 mmol Fe^(2+)/g降低为0.237 mmol Fe^(2+)/g。因此,梁王茶多糖可作为一种天然活性多糖具有潜在的开发价值和深入研究的意义。To purifiy and evaluate antioxidant activity of polysaccharide from nothopanax delavayi,Sevag method,AB-8 macroporous resin,DEAE-52 cellulose column,and gel were used to purify rude polysaccharide of nothopanax delavayi.Their polysaccharides,uronic acids,protein,total polyphenols,and total flavonoids contents were measured.Then their structural characteristics were characterized by fourier transform infrared spectroscopy(FT-IR)and scanning electron microscope(SEM).Moreover,their antioxidant capacities were evaluated by DPPH radical scavenging assay and FRAP method.The results showed that purified polysaccharides of nothopanax delavayi had a clear hollow spheroid structure,and the purification of rude polysaccharides was effective.The results of antioxidant assays showed that polysaccharides of nothopanax delavayi had certain antioxidant activities.And the DPPH radical scavenging capacity of purified polysaccharide was higher than rude polysaccharide,the values of IC50 changed from 7.274 mg·mL^(-1)to 7.101 mg·mL^(-1).While the ferric reducing antioxidant power were lower than rude polysaccharide,the antioxidant activities changed from 0.342 mmol Fe^(2+)/g to 0.237 mmol Fe^(2+)/g.Therefore,nothopanax delavayi polysaccharides could be used as the source of natural active polysaccharide,which had the potential development value and the significance of further study.
