摘  要:目的探讨长链非编码核糖核酸ZNRD1-AS1(lncRNA ZNRD1-AS1)在胃癌组织中的表达特点及其诊断学价值。方法选取嘉兴学院附属第二医院胃肠外科收治的96例经手术治疗、且留有手术样本的胃癌患者作为研究对象,根据是否发生转移分为转移癌41例和原位癌55例。详细统计患者的临床病理和预后资料,并采用荧光定量聚合酶链反应(PCR)实验检测癌组织及癌旁健康组织的lncRNA ZNRD1-AS1和微小核糖核酸-375模拟物(miR-375 mimic)表达水平。两组之间的比较行配对设计或独立样本t检验,多组间的比较行单因素方差分析。结果癌组织的lncRNA ZNRD1-AS1表达水平高于癌旁健康组织(1.051±0.205比0.645±0.145,t=15.845,P<0.001);癌组织的miR-375表达水平低于癌旁健康组织(1.014±0.197比1.365±0.245,t=10.965,P<0.001)。转移癌组织的lncRNA ZNRD1-AS1表达水平高于原位癌组织(1.201±0.165比0.953±0.148,t=3.604,P<0.01);转移癌组织的miR-375表达水平低于原位癌组织(0.995±0.164比1.193±0.176,t=5.612,P<0.001)。不同TNM分期癌组织的lncRNA ZNRD1-AS1表达水平也不同(F=14.437,P<0.001),Ⅰ期低于Ⅱ期(0.937±0.157比1.095±0.187,P<0.001);Ⅱ期低于Ⅲ期(1.095±0.187比1.185±0.154,P<0.001)。根据lncRNA ZNRD1-AS1表达水平的中位数把患者分为高表达组48例和低表达组48例,高表达组的总生存率低于低表达组[52.1%(25/48)比72.9%(35/48),χ^(2)=4.444,P<0.05],无进展生存率也低于低表达组[41.7%(20/48)比64.5%(31/48),χ^(2)=5.061,P<0.05]。结论lncRNA ZNRD1-AS1与胃癌的发生与发展密切相关。Objective To investigate the expression characteristics and diagnostic value of long-stranded non-coding RNA ZNRD1-AS1(lncRNA ZNRDI-ASI)in stomach cancer.Methods Totally,96 patients with gastric cancer who underwent surgery and retained surgical samples in the department of gastrointestinal surgery of the second hospital of Jiaxing university were selected and classified as 41 cases of metastatic cancer and 55 cases of in situ cancer according to whether there was metastasis.The detailed information of their clinicopathological and prognostic data were recorded,and the expression levels of ln-cRNA ZNRD1-AS1 and miR-375 in both cancerous tissues and adjacent healthy tissues were detected by fluorescence quantification polymerase chain reaction(PCR).Results The expression level of lncRNA ZNRD1-AS1 in cancer tissues was higher than that in adjacent healthy tissues(1.051±0.205 vs.0.645±0.145,t=15.845,P<0.001);and the expression level of miR-375 in cancerous tissues was lower than that of(1.014±0.197 vs.1.365±0.245,t=10.965,P<0.001).The lncRNA ZNRD1-AS1 expression levels in metastatic carcinoma tissues were higher than those in situ carcinoma tissues(1.201±0.165 vs.0.953±0.148,t=3.604,P<0.01);and the expression level of miR-375 in metastatic tissues was lower than that of carcinoma-in-situ tissues(0.995±0.164 vs.1.193±0.176,t=5.612,P<0.001).The expression lev-els of lncRNA ZNRDI-ASI in different TNM stage cancer tissues were also different(F=14.437,P<0.001),those in stage Ⅰ were lower than stage Ⅱ(0.937±0.157 vs.1.095±0.187,P<0.001);those in stage Ⅱ were lower than stage Ⅱ(1.095±0.187 vs.1.185±0.154,P<0.001).According to the median expression level of lncRNA ZNRDI-ASI,the patients were divided into 48 cases in the high-expression group and 48 cases in the low-expression group,and the overall survival rate of the high-expression group was lower than that of the low-expression group[52.1%(25/48)vs.72.9%(35/48),χ^(2)=4.444,P<0.05],the progression-free survival rate of the high-expression group was al
