摘 要:目的探讨食管鳞状细胞癌(ESCC)组织中Krüppel样因子4(KLF4)和Yes相关蛋白1(YAP1)的表达及临床意义。方法收集2017年5月至2022年12月大庆龙南医院(齐齐哈尔医学院第五附属医院)和大庆油田总医院病理学检查确诊的69例ESCC患者的癌组织和对应癌旁组织标本,采用免疫组织化学法检测上述组织中KLF4和YAP1表达水平,进行χ^(2)检验,结合临床资料分析两者的表达与ESCC临床病理特征的关系。组间比较采用χ^(2)=检验。结果ESCC癌组织中KLF4阳性表达率46.38%(32/69),明显低于对应癌旁组织中KLF4阳性表达率81.16%(56/69),差异有统计学意义(χ^(2)=18.066,P<0.01)。KLF4表达水平与ESCC患者淋巴结转移、TNM分期(TNM stage)明显相关(χ^(2)=4.512、5.301,P<0.05),KLF4表达水平与ESCC患者性别、年龄、组织学分化程度无明显相关(χ^(2)=0.002、0.006、0.165,P>0.05)。ESCC癌组织中YAP1阳性表达率63.77%(44/69),明显高于对应癌旁组织中YAP1阳性表达率15.94%(11/69),两者差异有统计学意义(χ^(2)=32.921,P<0.01)。YAP1表达水平与ESCC患者淋巴结转移、组织学分化程度、TNM分期明显相关(χ^(2)=6.287、4.540、4.681,P<0.05),YAP1表达水平与ESCC患者性别、年龄无明显相关(χ^(2)=0.0040.042,P>0.05)。结论ESCC癌组织中,KLF4表达下调和YAP1表达上调可能与ESCC进展和不良临床病理特征有关,检测KLF4和YAP1有助于判断ESCC生物学行为。Objective To investigate the expression and clinical significance of Kruppel-like factor 4(KLF4)and Yes-associated protein 1(YAPI)in esophageal squamous cell cancer(ESCC)tissues.Methods The cancer tissues and corresponding adjacent tissues of 69 ESCC patients confirmed by patho-logical examination in Daqing Longnan Hospital(the Fifth Affiliated Hospital of Qiqihar Medical College)and Daqing Oilfield General Hospital from May 2017 to December 2022 were collected.The expression levels of KLF4 and YAP1 in the above tissues were detected by immunohistochemistry,and chi-square test was performed.The relationship between KLF4 and YAPI expression and clinicopathological characteristics of ESCC was analyzed in combination with clinical data.Results The positive expression rate of KLF4 in ESCC tissues was 46.38%(32/69),which was significantly lower than that in corresponding adjacent tis-sues[81.16%(56/69),χ^(2)=18.066,P<0.01].The expression level of KLF4 was significantly correla-ted with lymph node metastasis and tumor node metastasis(TNM)of ESCC patients(respectively,χ^(2)=4.512,5.301,P<0.05).The expression level of KLF4 was not significantly correlated with gender,age and histological differentiation of ESCC patients(respectively,χ^(2)=0.002,0.006,0.165,P>0.05).The positive expression rate of YAP1 in ESCC tissues was 63.77%(44/69),which was significantly high-er than that in corresponding adjacent tissues[15.94%(11/69),χ^(2)=32.921,P<0.01].The expression level of YAP1 was significantly correlated with lymph node metastasis,histological differentiation and TNM stage of ESCC patients(respectively,χ^(2)=6.287,4.540,4.681,P<0.05).The expression level of YAPI was not significantly correlated with gender and age of ESCC patients(respectively,χ^(2)=0.004,0.042,P>0.05).Conclusion Down-regulation of KLF4 and up-regulation of YAP1 in ESCC tissues may be related to the progression and adverse clinicopathological features of ESCC.Detection of KLF4 and YAP1 may help to determine the biological behavior of ESCC.
