摘 要:目的:探讨苦瓜皂苷对结肠癌细胞增殖及凋亡的影响及其可能作用机制。方法:体外培养结肠癌细胞Caco-2,分为对照组、苦瓜皂苷L组、苦瓜皂苷M组、苦瓜皂苷H组、si-NC组、si-circ_0008274组、苦瓜皂苷H+pcDNA-circ_0008274组;MTT法、平板克隆实验、流式细胞术分别检测细胞增殖、克隆形成及凋亡;qRT-PCR法检测circ_0008274、miR-377的表达量;双荧光素酶报告实验检测circ_0008274与miR-377的靶向关系。结果:与对照组比较,苦瓜皂苷L组、苦瓜皂苷M组、苦瓜皂苷H组细胞增殖抑制率、miR-377的表达量、细胞凋亡率升高(P<0.05),克隆形成数减少(P<0.05),circ_0008274的表达量降低(P<0.05);与si-NC组比较,si-circ_0008274组细胞增殖抑制率、细胞凋亡率升高(P<0.05),克隆形成数减少(P<0.05);circ_0008274可靶向调控miR-377的表达;与苦瓜皂苷H组比较,苦瓜皂苷H+pcDNA-circ_0008274组miR-377的表达量、细胞增殖抑制率、细胞凋亡率降低(P<0.05),克隆形成数增多(P<0.05)。结论:苦瓜皂苷可通过抑制circ_0008274表达而促进miR-377表达来诱导结肠癌细胞凋亡、抑制细胞增殖。Objective:To explore the effect of momordica charantia saponins on cell proliferation and apoptosis in colon cancer and its possible mechanism.Methods:Human colon cancer cells Caco-2 were cultured and divided into control group,momordica charantia saponinsL group,momordica charantia saponinsM group,momordica charantia saponinsH group,si-NC group,si-circ_0008274 group,momordica charantia saponinsH+pcDNA-circ_0008274 group.MTT method,plate colony assay and flow cytometry were performed to examine cell proliferation,colony formation and apoptosis.qRT-PCR method was used to detect the expression levels of circ_0008274 and miR-377.The dual luciferase reporter experiment was used to detect the targeting relationship between circ_0008274 and miR-377.Results:Compared with the control group,the inhibition rate,the expression of miR-377,cell apoptosis rate in the momordica charantia saponinsL group,momordica charantia saponinsM group,momordica charantia saponinsH group were increased(P<0.05),while the number of clone formation was decreased(P<0.05),and the expression of circ_0008274 was decreased(P<0.05).Compared with the si-NC group,the inhibition rate,apoptosis rate in the si-circ_0008274 group were increased(P<0.05),while the number of clone formation was decreased(P<0.05).circ_0008274 could target and regulate the expression of miR-377.Compared with momordica charantia saponinsH group,the expression of miR-377,cell proliferation inhibition rate,cell apoptosis rate in the momordica charantia saponinsH+pcDNA-circ_0008274 group were decreased(P<0.05),while the number of clone formation was increased(P<0.05).Conclusion:Momordica charantia saponins could promote the expression of miR-377 by inhibiting the expression of circ_0008274,thereby inducing colon cancer apoptosis and inhibiting cell proliferation.
