摘　　要：目的:探究泛素结合酶E2C(UBE2C)表达对肝内胆管癌(iCCA)细胞增殖与侵袭的影响及其预后评估价值。方法:收集12例iCCA患者肿瘤组织和癌旁正常组织,检测UBE2C转录水平。选取58例根治性切除术后iCCA患者的肿瘤组织标本,通过免疫组化检测UBE2C的表达情况,并分析其与患者临床病理特征及预后的关系。使用siRNA沉默RBE细胞中UBE2C的表达,通过MTT与Transwell实验检测UBE2C对细胞增殖及侵袭的影响。结果:iCCA组织中UBE2C表达上调,其表达与低分化、淋巴结转移、术后复发以及TNM分期等临床病理特征密切相关,UBE2C阳性患者预后明显不良。细胞学实验发现沉默UBE2C后,iCCA细胞的增殖及侵袭能力均下降。结论:UBE2C表达具有评估iCCA预后的价值,其能够诱导肿瘤细胞的增殖侵袭从而发挥促癌作用,有可能成为iCCA治疗的潜在分子靶点。Objective:To investigate the expression of ubiquitin-conjugating enzyme E2C(UBE2C)in intrahepatic cholangiocarcinoma(iCCA)tissues and its correlation with prognosis,and to explore the effect of UBE2C on the proliferation and invasion of cholangiocarcinoma cells.Methods:12 patients with iCCA were selected to analyse the expression of UBE2C in the cancerous tissues and paired adjacent normal tissues.Meanwhile,58 tumor samples from patients with iCCA that received radical surgery were enrolled to explore the relationship between UBE2C expression and clinicopathological parameters.UBE2C was knockdown by siRNA,and subsequently MTT and Transwell assay were applied to investigate cell proliferation and invasion.Results:The expression of UBE2C was upregulated in iCCA tissues compared to those in adjacent normal tissues.Expression of UBE2C was associated with poor differentiation,lymph node metastasis,post-operative recurrence and advanced TNM stage.In addtion,patients with high UBE2C expression suffered a poorer prognosis.In vitro study indicated that the proliferation and invasion capability of RBE cells was significantly inhibited upon knockdown of UBE2C.Conclusions:As an indicator of poor prognosis,UBE2C promotes iCCA progression by promoting tumor proliferation and invation,wich might act as a potentially new therapeutic target of iCCA.
