摘 要:目的对比不同制备方法对去卵巢小鼠模型的影响。方法结扎法、电烙法完全摘除小鼠双侧卵巢,检测阴道涂片、脏器湿重、生化指标,Micro-CT检测股骨组织形态变化,HE染色观察股骨、子宫、胸腺、脾脏病理变化,对比两种不同方法对去卵巢小鼠模型的影响。结果与假手术组相比,结扎法、电烙法均可使去卵巢小鼠连续处于动情间期,子宫、脾脏、胸腺脏器湿重降低,血清中骨特异性碱性磷酸酶(bone-specific alkaline phosphatase,BALP)、骨钙素(bone gla protein,BGP)水平降低;血清性激素水平紊乱,雌二醇(estradiol,E_(2))水平降低、促卵泡生成素(follicle stimulating hormone,FSH)水平升高;小鼠股骨骨小梁厚度(trabecular thickness,Tb.Th)、骨矿物质密度(bonemineral density,Tb.BMD)、连接密度(connectivity density,Conn.D)水平降低,骨小梁分离度(trabecular spacing,Tb.Sp)水平升高,皮质骨骨矿物质密度(bone mineral density,Ct.BMD)、皮质骨厚度(cortical bone thickness,Ct.Th)水平降低;去卵巢小鼠胸腺皮质厚度、脾小结体积明显减小,子宫出现明显萎缩,股骨骨小梁排列紊乱,骨小梁结构变细,破骨细胞数量明显增多。结论结扎法、电烙法均能成功制备去卵巢小鼠模型,从制备方法上看电烙法更简便,为卵巢摘除法相关研究提供实验基础。Aim To compare the effects of different methods on the preparation of ovariectomized mouse models.Methods The bilateral ovaries of mouse were completely removed by desmurgia and diathermocoagulation respectively.The effects of desmurgia and diathermocoagulation methods on ovariectomized mouse models were compared by detecting vaginal smears,organ indexes,biochemical indexes,Micro-CT was used to detect the morphological changes in femur tissue,and HE staining was used to observe the pathological changes in femur,uterus,thymus and spleen.Results Compared with the control group,the estrous cycle of mouse was disordered by desmurgia and diathermocoagulation,the indexes of uterus,spleen and thymus were reduced,the levels of BGP,BALP and E_(2) in serum decreased,the levels of FSH increased,and the levels of TB.Th,TB.BMD and Conn.D decreased,the level of TB.Sp increased,and the level of CT.BMD and CT.Th decreased.HE staining showed the thickness of the thymic cortex and the volume of the spleen nodula were significantly reduced,the uterine volume was atrophy significant,the trabecular bone arrangement was disordered,the structure of the bone became thinner,and the number of osteoclasts increased.Conclusions Ovariectomized mouse model is successfully prepared by desmurgia and diathermocoagulation,and the diathermocoagulation method is more convenient,and it provides an experimental basis for the research related to ovarian removal.
