摘 要:目的:分析CDGSH铁硫结构域2(CDGSH iron sulfur domain 2,CISD2)在胃腺癌组织中的表达及其与血管内皮生长因子(vascular endothelial growth factor,VEGF)和肿瘤微血管密度(microvessel density,MVD)的相关性及临床意义。方法:收集胃腺癌组织(胃癌组)及配对的癌旁组织(癌旁组)各357例,免疫组化法检测CISD2、VEGF的表达,CD34标记血管内皮细胞并计算MVD值。分析CISD2、VEGF的表达及其与临床病理参数的相关性。分析MVD、CISD2和VEGF的两两相关性。结果:胃癌组中,CISD2和VEGF的表达均高于癌旁组(70.87%比25.77%,61.34%比19.89%,均P<0.001)。CISD2、VEGF的表达与胃腺癌的浸润深度、分化程度、淋巴结转移、脉管及神经侵犯、肿瘤的TNM分期均相关(均P<0.05)。胃癌组中,CISD2阳性者的MVD值高于CISD2阴性者(35.67±6.06比31.98±4.87,P<0.001),VEGF阳性者的MVD值高于VEGF阴性者(36.64±6.08比31.38±4.54,P<0.001);癌旁组中亦然。胃癌组中,CISD2与VEGF的表达呈正相关(r=0.706,P<0.001)。结论:CISD2在胃腺癌中高表达,且与组织学分化、浸润、转移及高微血管密度等相关。CISD2可能与VEGF共同参与胃腺癌组织中微血管的形成。Objective:To investigate the expression and clinical significance of CDGSH iron sulfur domain 2(CISD2)in gastric adenocarcinoma,and its correlation with vascular endothelial growth factor(VEGF)and tumor microvessel density(MVD).Methods:Gastric adenocarcinoma tissues(cancer group,n=357)and their paired adjacent normal gastric mucosa tissues(control group,n=357)were included in this study.The expression of CISD2 and VEGF was detected by immunohistochemical stain.Vascular endothelial cells were labeled with CD34,and MVD was calculated.The expression of CISD2 and VEGF and its correlation with clinicopathological parameters was analyzed.The correlation between MVD,CISD2 and VEGF was analyzed.Results:The expression of CISD2 in the cancer group was higher than that in the control group(70.87%vs 25.77%,P<0.001).The expression of VEGF in the cancer group was higher than that in the control group(61.34%vs 19.89%,P<0.001).The expression of CISD2 and VEGF correlated with the depth of invasion,the grade of differentiation,lymph node metastasis,vascular and neural invasion and TNM stage of gastric adenocarcinoma(all P<0.05).In the cancer group,CISD2 positive patients had higher MVD than CISD2 negative patients(35.67±6.06 vs 31.98±4.87,P<0.001).VEGF positive patients also had higher MVD than VEGF negative patients in the cancer group(36.64±6.08 vs 31.38±4.54,P<0.001).In the cancer group,a positive correlation between the expression of CISD2 and the expression of VEGF was observed(r=0.706,P<0.001).Conclusion:CISD2 is overexpressed in gastric adenocarcinoma compared to adjacent normal gastric mucosa.The high expression of CISD2 correlated with histological differentiation,invasion and metastasis of gastric adenocarcinoma.CISD2 may be involved in the formation of microvessels in gastric adenocarcinoma.VEGF may play an important role with CISD2 in the formation of microvessels.
