摘  要:为了研究多穗柯(Lithocarpus polystachyus Rehd)MYB转录因子在光胁迫时调控多穗柯黄酮类物质合成的作用,以不同光处理条件的多穗柯为试验材料,通过转录组学测序和生物信息学分析方法,对多穗柯MYB转录因子和多穗柯黄酮类物质合成关键基因进行了分析。结果标明:在多穗柯的转录组学测序数据中,鉴定得到60个LpMYB转录因子,按照DNA结合结构域的特点,将这些转录因子分为4个亚族(1R-MYB,R2R3-MYB,3R-MYB,4R-MYB),其中R2R3-MYB数量最多,占总LpMYB转录因子的43.33%。多穗柯大部分MYB转录因子能够适应红光胁迫;长时间的光照、适当的光照度提高了LpMYB转录因子的表达。鉴定得到10个参与调控黄酮类物质生物合成的S4、S5、S6和S7亚族LpMYB转录因子。多穗柯黄酮类物质合成相关基因与LpMYB转录因子的相关性分析结果表明,LpMYB转录因子在调控黄酮化合物的合成中发挥着重要的作用,LpMYB转录因子能够正调控黄酮类物质合成基因PAL、C4H、CHS3、ANS的表达,从而提高多穗柯的品质。In order to investigate the role of MYB transcription factors in the regulation of flavonoid synthesis in Lithocarpus polystachyus Rehd under light stress,the key genes of MYB transcription factors and flavonoid synthesis in L.polystachyus were analyzed by transcriptomic sequencing and bioinformatics analysis using L.polystachyus as experimental material under different light treatment conditions.Sixty LpMYB transcription factors were identified in the transcriptomic sequencing data of L.polystachyus,and these transcription factors were classified into four subgroups(1R-MYB,R2R3-MYB,3R-MYB and 4R-MYB)according to the characteristics of the DNA-binding structural domains,among which R2R3-MYB was the most abundant,accounting for 43.33%of the total LpMYB transcription factors.The majority of MYB transcription factors in L.polystachyus were able to adapt to red light stress;prolonged light exposure and appropriate light intensity increased the expression of LpMYB transcription factors.Ten LpMYB transcription factors from subfamilies of S4,S5,S6 and S7 involved in the regulation of flavonoid biosynthesis were identified.The correlation analysis between the genes related to flavonoid synthesis and LpMYB transcription factors of L.polystachyus showed that LpMYB transcription factors play an important role in regulating the synthesis of flavonoids,and LpMYB transcription factors can positively regulate the expression of flavonoid synthesis genes PAL,C4H,CHS3 and ANS,thus improving the quality of L.polystachyus.
