摘  要:目的探究尾穗苋Amaranthus caudatus L.化学成分及其抗氧化活性。方法尾穗苋70%乙醇提取物乙酸乙酯部分采用硅胶、葡聚糖凝胶LH-20、HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用还原性、清除DPPH、清除羟自由基、抑制脱氧核糖氧化能力评价其抗氧化活性。结果从中分离得到10个化合物,分别鉴定为十六烷酸(1)、α-菠甾醇(2)、α-菠甾醇β-D-吡喃葡萄糖苷(3)、尿嘧啶(4)、反式阿魏酸(5)、槲皮素-3-芸香糖苷(6)、3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl-2β,3β,6β,23-tetrahydroxyolean-12-en-28-oic acid 28-O-β-D-glucopyranoside(7)、3-O-β-D-glucopyranosyl-2β,3β-dihydroxy-30-noroleane-12,20(29)-diene-23,28-dioic acid 28-O-β-D-glucopyranosyl ester(8)、3,4,5-trihydroxydehydro-α-ionol-9-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(9)、3,4,5-trihydroxydehydro-α-ionol-9-β-D-glucopyranoside(10)。化合物5~6对·DPPH的IC 50值分别为(0.598±0.088)、(0.405±0.043)mg/mL。结论化合物1~6为首次从该植物中分离到。化合物5~6具有较强的抗氧化活性。AIM To study the chemical constituents from Amaranthus caudatus L.and their antioxidation activities.METHODS The ethyl acetate fraction of the 70%ethanol extract from A.caudatus was isolated and purified by silica gel,Sephadex gel LH-20 and HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their antioxidant activities were tested by reducing activity,scavenging DPPH,scavenging hydroxyl radicals and inhibiting deoxyribose oxidation.RESULTS Ten compounds were isolated and identified as hexadecanoic acid(1),α-spinasteryl(2),α-spinasterylβ-D-glucopyranoside(3),uracil(4),trans-ferulic acid(5),quercetin-3-rutinoside(6),3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl-2β,3β,6β,23-tetrahydroxyolean-12-en-28-oic acid 28-O-β-D-glucopyranoside(7),3-O-β-D-glucopyranosyl-2β,3β-dihydroxy-30-noroleane-12,20(29)-diene-23,28-dioic acid 28-O-β-D-glucopyranosyl ester(8),3,4,5-trihydroxydehydro-α-ionol-9-O-α-L-rhamnopyranosyl-(1→6)-β-D-glucopyranoside(9),and 3,4,5-trihydroxydehydro-α-ionol-9-β-D-glucopyranoside(10).The IC 50 values of compounds 5-6 on DPPH·were(0.598±0.088)and(0.405±0.043)mg/mL.CONCLUSION Compounds 1-6 are isolated from A.caudatus for the first time.Compounds 5-6 have significant antioxidant activities.
