摘  要:目的探讨大蒜素对高糖诱导的H9c2心肌细胞损伤、炎性反应以及信号转导和转录激活因子3(STAT3)/缺氧诱导因子1α(HIF-1α)通路的影响。方法体外培养大鼠H9c2心肌细胞,分为对照组、高糖组、大蒜素低、中、高剂量组。噻唑蓝法检测H9c2细胞活力;流式细胞仪检测H9c2细胞凋亡率;比色法检测H9c2细胞中活性氧水平;酶联免疫吸附法检测H9c2细胞中白细胞介素6(IL-6)和TNF-α水平;荧光定量PCR法检测H9c2细胞中STAT3、HIF-1αmRNA表达;蛋白印迹法检测H9c2细胞中STAT3、HIF-1α蛋白表达。结果与对照组比较,高糖组细胞活力明显降低,凋亡率、活性氧、IL-6、TNF-α水平、STAT3 mRNA、HIF-1αmRNA、STAT3蛋白、HIF-1α蛋白表达明显升高(P<0.05);与高糖组比较,大蒜素低、中、高剂量组细胞活力明显升高[(65.48±7.54)%、(77.31±7.82)%、(89.26±8.64)%vs(54.15±6.03)%,P<0.05],凋亡率[(26.47±3.76)%、(20.19±3.05)%、(12.57±2.24)%vs(31.80±4.23)%]、活性氧[(264.51±30.26)%、(227.64±27.77)%、(158.29±19.50)%vs(359.26±47.61)%]、IL-6[(74.64±5.87)ng/L、(61.58±5.11)ng/L、(50.72±3.60)ng/L vs(85.61±7.43)ng/L]、TNF-α[(54.31±4.62)ng/L、(42.35±4.11)ng/L、(27.59±3.51)ng/L vs(68.24±6.64)ng/L]、STAT3 mRNA(2.72±0.34、2.15±0.27、1.48±0.24 vs 3.32±0.41)、HIF-1αmRNA(3.46±0.39、2.85±0.22、2.14±0.18 vs 4.35±0.45)、STAT3蛋白(1.12±0.13、0.90±0.08、0.52±0.05 vs 1.53±0.17)、HIF-1α蛋白(1.13±0.11、0.77±0.07、0.41±0.06 vs 1.60±0.15)表达显著降低,差异有统计学意义(P<0.05)。结论大蒜素可减轻高糖诱导的H9c2心肌细胞损伤和炎性反应,可能与抑制STAT3/HIF-1α通路激活有关。Objective To investigate the effect of allicin on high glucose-induced injury,inflammatory response and signal transducer and activator of transcription 3(STAT3)/hypoxia inducible factor-1α(HIF-1α)pathway in H9c2 cells.Methods H9c2 cells were divided into control group,high glucose group,and allicin low-,medium-and high-dose groups.Methyl thiazolyl tetrazolium assay was used to detect cell viability;flow cytometry was employed to detect the apoptotic rate;colorimetry was adopted to measure the level of reactive oxygen species(ROS);ELISA was conducted to test the levels of IL-6 and TNF-α;fluorescence quantitative PCR and Western blotting were used to detect the expression of STAT3 and HIF-1α at the mRNA and protein levels respectively.Results Compared with the control group,the cell vitality was significantly decreased,and the apoptotic rate,ROS,IL-6 and TNF-α levels,and mRNA and protein levels of STAT3 and HIF-1α were significantly higher in the high glucose group(P<0.05).Compared with the high glucose group,the cell viability[(65.48±7.54)%,(77.31±7.82)%,(89.26±8.64)%vs(54.15±6.03)%]in allicin low,medium and high dose groups was significantly increased,apoptotic rate[(26.47±3.76)%,(20.19±3.05)%,(12.57±2.24)%vs(31.80±4.23)%],ROS[(264.51±30.26)%,(227.64±27.77)%,(158.29±19.50)%vs(359.26±47.61)%],IL-6[(74.64±5.87)ng/L,(61.58±5.11)ng/L,(50.72±3.60)ng/L vs(85.61±7.43)ng/L],TNF-α[(54.31±4.62)ng/L,(42.35±4.11)ng/L,(27.59±3.51)ng/L vs(68.24±6.64)ng/L],STAT3 mRNA(2.72±0.34,2.15±0.27,1.48±0.24 vs 3.32±0.41),HIF-1α mRNA(3.46±0.39,2.85±0.22,2.14±0.18 vs 4.35±0.45),STAT3 protein(1.12±0.13,0.90±0.08,0.52±0.05 vs 1.53±0.17),and HIF-1α protein(1.13±0.11,0.77±0.07,0.41±0.06 vs 1.60±0.15)expression was significantly decreased(P<0.05).Conclusion Allicin alleviates high glucose induced injury and inflammatory response in H9c2 cells,which may be related to its inhibiting the activation of STAT3/HIF-1α pathway.
