摘  要:山楂(Crataegus pinnatifida Bunge)提取物牡荆素鼠李糖苷(vitexin-2"-O-rhamnoside,VOR)是一种黄酮苷类化合物,具有心脏保护作用。本实验旨在考察牡荆素鼠李糖苷对力竭训练大鼠心肌损伤的保护作用。本实验通过力竭游泳建立了力竭训练大鼠模型,训练当天对大鼠分别灌胃3个剂量(20,40和80 mg/kg)的牡荆素鼠李糖苷,共给药4周。然后检测了大鼠的力竭时间、心肌损伤指标(LDH,CK和cTnI)、心肌形态、心肌氧化应激(SOD,CAT和MDA)和炎症(IL-1β,IL-6和TNF-α)指标。通过Western blot检测心肌组织中Nrf2、Keap1、HO-1、NQO-1、p38 MAPK、p-p38 MAPK、NF-κB p65、p-NF-κB p65蛋白的表达水平。结果表明,3种剂量的牡荆素鼠李糖苷均提高了大鼠的力竭时间,降低了血清LDH、CK和cTnI水平,升高了心肌组织SOD和CAT水平,降低了MDA水平,降低了心肌组织IL-1β、IL-6和TNF-α水平,升高了心肌组织细胞核Nrf2、HO-1和NQO-1蛋白相对表达量,降低了心肌组织Keap1蛋白相对表达量,降低了心肌组织p38 MAPK和NF-κB p65磷酸化水平(P<0.05)。本实验表明牡荆素鼠李糖苷对力竭训练大鼠具有心肌保护作用,其机制涉及激活Nrf2信号转导途径和失活p38 MAPK/NF-κB信号转导途径。Vitexin-2"-O-rhamnoside(VOR)is a flavonoid glycoside from Crataegus pinnatifida Bunge,which has cardioprotective effect.The purpose of this study was to investigate the protective effect of vitexin-2"-O-rhamnoside on myocardial injury in exhaustive training rats.In this experiment,the rat model of exhaustive training was established by exhaustive swimming.On the training day,the rats were given vitexin-2"-O-rhamnoside of three doses(20,40 and 80 mg/kg)respectively for 4 weeks.Then the exhaustion time,myocardial injury indexes(LDH,CK and cTnI),myocardial morphology,myocardial oxidative stress(SOD,CAT and MDA)and inflammation(IL-1β,IL-6 and TNF-α)were measured.The expression levels of Nrf2,Keap1,HO-1,NQO-1,p38 MAPK,p-p38 MAPK,NF-κB p65 and p-NF-κB p65 protein in myocardial tissue were detected by Western blot.The results showed that three doses of vitexin-2"-O-rhamnoside increased the exhaustion time,decreased the levels of serum LDH,CK and cTnI,increased the levels of SOD and CAT in myocardial tissue,decreased the level of MDA in myocardial tissue,decreased the levels of IL-1β,IL-6 and TNF-αin myocardial tissue,increased the relative expression of nuclear Nrf2,HO-1 and NQO-1 protein in myocardial tissue,decreased the relative expression of Keap1 protein in myocardial tissue,decreased the phosphorylation levels of p38 MAPK and NF-κB p65 in myocardial tissue(P<0.05).This study shows that vitexin-2"-O-rhamnoside has a myocardial protective effect on exhaustive training rats,and its mechanism involves activating Nrf2 signal transduction pathway and inactivating p38 MAPK and NF-κB signal transduction pathway.
