摘 要:目的:研究柴达木黑果枸杞花青素对中波紫外线(UVB)照射后体外培养人皮肤成纤维细胞(HSF)衰老的保护作用及衰老相关基因p53、p21、微RNA(miR)-34c和沉默信息相关调节因子(SIRT)1 mRNA表达水平的影响。方法:取对数生长期的HSF,分为对照组、UVB照射组,柴达木黑果枸杞花青素低剂量组(0.1 g/L)、柴达木黑果枸杞花青素中剂量组(0.5 g/L)和柴达木黑果枸杞花青素高剂量组(1.0 g/L)。UVB照射前24 h用不同浓度的柴达木黑果枸杞花青素预处理HSF。UVB照射后72 h,β-半乳糖苷酶(SA-β-gal)染色法检测衰老细胞比例,定量反转录聚合酶链反应(qRT-PCR)法检测细胞中衰老相关基因p53、p21、miR-34c及SIRT1 mRNA的表达水平。结果:与对照组相比,UVB照射组细胞SA-β-gal染色阳性率及p53、p21、miR-34c和SIRT1 mRNA表达水平均升高(P<0.05)。与UVB照射组相比,柴达木黑果枸杞花青素(0.1 g/L、0.5 g/L和1.0 g/L)组细胞SA-β-gal染色阳性率,p53、p21、miR-34c及SIRT1 mRNA的表达水平,均呈剂量依赖性降低(P<0.05)。结论:黑果枸杞花青素可能通过下调p53/miR-34c正反馈调节来减轻HSF的照射损伤,进而发挥抗衰老作用。Objective:To study the effects of anthocyanins in Lycium ruthenicum,produced in Chaidamu,on medium-wave ultraviolet(UVB)radiation damage in vitro-cultured human skin fibroblasts(HSFs).Methods:HSFs in the logarithmic growth phase were divided into control group,UVB radiation group,low-(0.1 g/L),medium-(0.5 g/L)and high-dose(1.0 g/L)anthocyanin groups.HSFs were pretreated with different concentrations of anthocyanin in Lycium ruthenicum from Chaidamu at 24h before UVB radiation.At 72h after UVB irradiation,the proportion of senescent cells was detected by SA-β-gal staining,and the ex-pression levels of p53,p21,miR-34c and SIRTI mRNA were detected by qRT-PCR.Results:Compared with those in the control group,the positive rate of SA-β-gal staining and the expression levels of p53,p21,miR-34c and SIRTI mRNA in the UVB radiation group increased(P<0.05).Compared with those in the UVB radiation group,the positive rates of SA-β-gal staining in three anthocyanin groups reduced,and the expression levels of p53,p21,miR-34c and SIRTI mRNA decreased(P<0.05),the positive rate of SA-β-gal staining,p53,p21,and miR-34c mRNA expression in cells were dose-dependent.Conclusion:Anthocyanins in Lycium ruthenicum,produced in Chaidamu,may reduce radiation damage on HSFs by down-regulating the positive feedback regulation of p53/miR-34.
