摘  要:为了分析冷鲜肉在冷冻过程中微生物群落变化,选取不同冷冻时间的冷鲜肉作为样品,采用Illumina MiSeq技术解析微生物结构,并获取微生物群落多样性分析结果;随着冷鲜肉冷藏时间的增长,样品OTU数量从209下降至98~125。通过分析Chao index与Shannon index的计算结果可知,冷冻时间较短的试验D组具有更高值。冷鲜肉微生物群落结构中,样品中嗜冷菌属和Oceanisphaera的相对丰度超过89%。微生物群落主成分分析时,冷冻时间较短的样本点与冷冻时间最长的样本点,在PC1水平上处于两个极端,菌群结构变化显著。随着冷鲜肉冷冻处理时间的增长,食品微生物群落多样性大幅下降,但是嗜冷菌属与Oceanisphaera作为优势菌数量开始增加。结果表明,在冷鲜肉食品微生物检测过程中,该方法可以获得更加全面的微生物群落多样性分析结果,快速得出冷鲜肉处理过程中影响较大的优势腐败菌,为后期食品质量提升提供依据。In order to analyze the changes of microbial community during freezing of cold fresh meat,cold fresh meat with different freezing time was selected as samples,and the microbial structure was analyzed by Illumina MiSeq technology,and the results of microbial community diversity analysis were obtained.With the increase of cold storage time,the number of OTU samples decreased from 209 to 98-125.By analyzing the calculation results of the Chao index and Shannon index,it can be seen that group D with shorter freezing time has higher value.In the microbial community structure of chilled meat,the relative abundance of psychrophilic bacteria and Oceanisphaera was more than 89%.In principal component analysis of microbial community,the samples with shorter freezing time and the samples with longest freezing time were in two extremes at PC1 level,and the change of microbial community structure was significant.With the increase of freezing time,the diversity of food microbial community decreased significantly,but the number of psychrophilic bacteria and Oceanisphaera as dominant bacteria began to increase.The results show that in the process of microbial detection of cold and fresh meat food,this method can obtain more comprehensive microbial community diversity analysis results,quickly obtain the dominant spoilage bacteria with great influence in the process of cold and fresh meat treatment,and provide a basis for the later improvement of food quality.
