摘  要:目的探讨circ_0000003对皮肤鳞状细胞癌(cSCC)细胞生物学行为的影响及其可能作用机制。方法应用实时定量聚合酶链反应(qRT-PCR)法检测正常皮肤组织、cSCC组织与cSCC细胞SCL-I中circ_0000003、微小核糖核酸-1238(miR-1238)的表达量;通过干扰SCL-I细胞circ_0000003表达、上调miR-1238的表达、抑制miR-1238的表达,将SCL-I细胞分为si-NC组、si-circ_0000003组、miR-NC组、miR-1238组、si-circ_0000003+anti-miR-NC组、sicirc_0000003+anti-miR-1238组;甲基噻唑基四唑(MTT)法与平板克隆形成实验检测细胞活力、集落形成数;Transwell实验检测细胞迁移及侵袭能力;双荧光素酶报告实验检测circ_0000003和miR-1238的靶向关系。结果与正常皮肤组织比较,cSCC组织中circ_0000003的表达量升高(P<0.05),miR-1238的表达量降低(P<0.05);与si-NC组比较,si-circ_0000003组细胞活力降低(P<0.05),集落形成数、迁移及侵袭细胞数减少(P<0.05);与miR-NC组比较,miR-1238组细胞活力降低(P<0.05),集落形成数、迁移及侵袭细胞数减少(P<0.05);circ_0000003可靶向调控miR-1238;与si-circ_0000003+anti-miR-NC组比较,si-circ_0000003+anti-miR-1238组细胞活力升高(P<0.05),集落形成数、迁移及侵袭细胞数增多(P<0.05)。结论干扰circ_0000003表达可通过上调miR-1238表达而抑制cSCC细胞增殖、集落形成、迁移及侵袭。Objective In order to explore the effects of circ_0000003 on the biological behavior of cutaneous squamous cell carcinoma(cSCC)and its possible mechanism.Methods Quantitative real-time reverse transcription polymerase chain reaction(qRT-PCR)method was used to detect the expression of circ_0000003 and miR-1238 in normal skin tissue,cSCC tissue and human cSCC cell SCL-I.By interfering with the expression of circ_0000003,up-regulating the expression of microRNA-1238(miR-1238),and inhibiting the expression of miR-1238,SCL-I cells were divided into si-NC group,sicirc_0000003 group,miR-NC group,miR-1238 group,si-circ_0000003+anti-miR-NC group,si-circ_0000003+anti-miR-1238 group.Methylthiazolyl tetrazolium(MTT)method and plate colony formation experiment were used to detect cell viability and the number of colonies formed.Transwell test was used to detect cell migration and invasion ability.The dual luciferase reporter experiment was used to detect the targeting relationship between circ_0000003 and miR-1238.Results Compared with normal skin tissue,the expression of circ_0000003 in cSCC tissue was increased(P<0.05),while the expression of miR-1238 was decreased(P<0.05).Compared with the si-NC group,cell viability in the si-circ_0000003 group was decreased(P<0.05),and the number of colony formation,migration and invasion cells was decreased(P<0.05).Compared with the miR-NC group,the cell viability of the miR-1238 group was decreased(P<0.05),the number of colony formation,migration and invasion cells was decreased(P<0.05).circ_0000003 could target miR-1238.Compared with the si-circ_0000003+antimiR-NC group,the cell viability of the si-circ_0000003+anti-miR-1238 group was increased(P<0.05),and the number of colony formation,migration and invasion cells was increased(P<0.05).Conclusion Interference with the expression of circ_0000003 could inhibit the proliferation,colony formation,migration and invasion of cSCC cells by up-regulating the expression of miR-1238.
