摘 要:目的:研究柚皮素(NAR)对脑缺血再灌注(I/R)损伤神经保护作用的分子机制。方法:将大鼠原代皮层神经元分为正常组(NC)、模型组(OGD)、模型+柚皮苷组(OGD+NAR)和正常+NAR组(NC+NAR)。通过糖氧剥夺/再灌注(OGD/Rep)制备模型。噻唑蓝法(MTT)检测细胞增殖活力;流式细胞仪检测细胞活性氧(ROS)产生和凋亡变化;生化法检测抗氧化指标丙二醛(MDA)、谷胱甘肽(GSH)和超氧化物歧化酶(SOD)变化;Western blot检测自噬蛋白LC3、P62、抗细胞色素C抗体(Cytochrome C)、PTEN诱导假定激酶(PINK1)和Parkin表达变化。结果:NAR处理可增强模型组和正常培养的大鼠原代皮层神经细胞的增殖活力,抑制其凋亡和ROS产生。NAR可增强OGD/Rep模型的抗氧化能力,促进自噬蛋白LC3、PINK1和Parkin的表达,抑制P62和Cytochrome C表达。结论:柚皮素对脑I/R损伤具有保护作用,机制可能与清除ROS、提高抗氧化能力,促进细胞线粒体自噬有关。Objective:To explore the molecular mechanism of naringenin(NAR)in neuroprotective effect on cerebral ischemia reperfusion(I/R)injury.Methods:Rat primary cortical neurons were divided into normal group(NC),model group(OGD),model+naringin group(OGD+NAR)and normal+NAR group(NC+NAR).The model was established by glucose and oxygen deprivation/reperfusion(OGD/Rep).MTT assay was used to detect cell proliferation activity.ROS production and apoptosis were detected by flow cytometry.The changes of antioxidant indexes(MDA,GSH,SOD)were detected by biochemical method.Western blot was used to detect the expression of autophagy-related proteins LC3,P62,Cytochrome C,PINK1 and Parkin.Results:NAR treatment could enhance the proliferative activity of primary cortical neurons in model group and normal cultured rats cells,and inhibit their apoptosis and ROS production.NAR could enhance the antioxidant capacity of OGD/Rep model,promote the expression of autophagic proteins LC3,PINK1 and Parkin,and inhibit the expressions of P62 and Cytochrome C.Conclusion:Naringenin has protective effect on cerebral I/R injury,and its mechanism may be related to the elimination of ROS,improvement of antioxidant capacity,promoting of mitochondrial autophagy.
