摘 要:目的:探讨微小RNA-105-5p(miR-105-5p)靶向白细胞介素6受体(IL-6R)调控卵巢癌细胞(SKOV-3)增殖、迁移和侵袭。方法:实时定量PCR(RT-qPCR)分析miR-105-5p和IL-6R mRNA在卵巢癌组织中表达量。双荧光素酶报告实验确定miR-105-5p与IL-6R的靶向关系,RT-qPCR检测过表达miR-105-5p后SKOV-3细胞IL-6R表达量。细胞计数试剂盒(CCK-8)法和Transwell实验分析miR-105-5p和IL-6R表达对SKOV-3细胞增殖、迁移和侵袭的影响。结果:卵巢癌组织中miR-105-5p表达量较癌旁组织显著降低(P<0.05),而IL-6R mRNA表达量较癌旁组织显著升高(P<0.05)。IL-6R是miR-105-5p的靶基因,过表达miR-105-5p后IL-6R表达量显著降低(P<0.05)。过表达miR-105-5p或干扰IL-6R表达后SKOV-3细胞活力、迁移和侵袭细胞数显著降低(均P<0.05)。与过表达miR-105-5p比较,同时过表达miR-105-5p和IL-6R后SKOV-3细胞活力、迁移和侵袭细胞数显著升高(均P<0.05)。结论:miR-105-5p通过靶向IL-6R抑制卵巢癌细胞增殖和转移。Objective:To investigate whether microRNA-105-5p(miR-105-5p)targets the interleukin 6 receptor(IL-6R)to regulate the proliferation,migration and invasion of ovarian cancer cells(SKOV-3).Methods:The expressions of miR-105-5p and IL-6R mRNA in ovarian cancer tissues were analyzed by real-time quantitative PCR(RT-qPCR).The dual luciferase reporter experiment was used to confirm the targeting relationship between miR-105-5p and IL-6R,and RT-qPCR was used to detect the expression of IL-6R in SKOV-3 cells after miR-105-5p overexpression.The effects of miR-105-5p and IL-6R expression on the proliferation,migration and invasion of SKOV-3 cells were measured by cell counting kit(CCK-8)method and Transwell experiment.Results:The expression of miR-105-5p in ovarian cancer tissues was significantly lower than that in adjacent tissues(P<0.05),while the expression of IL-6R mRNA was significantly higher than that in adjacent tissues(P<0.05).IL-6R was the target gene of miR-105-5p.The expression of IL-6R was significantly decreased after miR-105-5p overexpression(P<0.05).After overexpression of miR-105-5p or interference with IL-6R expression,the cell viability,migration and invasion cells numbers of SKOV-3 were remarkably reduced(all P<0.05).Compared with overexpression of miR-105-5p,the cell viability,migration and invasion cell numbers of SKOV-3 were remarkably increased after overexpression of miR-105-5p and IL-6R at the same time(all P<0.05).Conclusion:MiR-105-5p can inhibit the proliferation and metastasis of ovarian cancer cells by targeting IL-6R.
