摘  要:目的探讨中药活血清解汤对重症急性胰腺炎(SAP)大鼠血红素加氧酶1(HO-1)及NOD样受体热蛋白结构域相关蛋白3(NLRP3)介导的细胞焦亡的调控作用。方法将60只SD雄性大鼠随机平均分为对照组、SAP模型组和活血清解汤组。SAP模型组及对照组采用经腹腔注射L-精氨酸复制SAP模型治疗,活血清解汤组采用活血清解汤治疗,对照组不做处理。比较各组血清HO-1及焦亡因子[NLPR3、白细胞介素-1β(IL-1β)]的变化及各组胰腺组织HO-1、NLPR3和IL-1β的基因及蛋白变化。结果3组血清HO-1水平组间、时间比较差异有统计学意义(P<0.05);3组血清HO-1水平交互比较差异无统计学意义。治疗后24、48、72 h,SAP模型组HO-1水平均高于对照组,差异有统计学意义(P<0.05);治疗后24、48、72 h,活血清解汤治疗组HO-1水平均高于SAP模型组及对照组,且SAP模型组高于对照组,差异有统计学意义(P<0.05)。3组IL-1β、IL-18水平组间、时间、交互比较差异有统计学意义(P<0.05)。治疗后24、48、72 h,SAP模型组大鼠血清、胰腺组织中IL-1β、IL-18水平均高于对照组和活血清解汤组,且活血清解汤组高于对照组(P<0.05)。SAP模型组大鼠血清、胰腺组织中HO-1、NLPR3和IL-1β水平均高于对照组(P<0.05)。活血清解汤组血清及胰腺组织中HO-1水平均高于对照组,NLPR3和IL-1β水平均低于对照组,差异有统计学意义(P<0.05)。结论中药活血清解汤可进一步提高SAP模型中HO-1水平,降低细胞焦亡因子NLRP3及IL-1β水平,减轻细胞损伤。Objective To investigate the regulatory effect of traditional Chinese medicine live serological decoction on cytopyroptosis mediated by heme oxygenase 1(HO-1)and NOD-like receptor thermoprotein domain-associated protein 3(NLRP3)in rats with severe acute pancreatitis(SAP).Methods 60 male rats with SD were randomly and evenly divided into control group,SAP model group and Huoxue qingjie decoction group.The SAP model group and the control group were treated with intraperitoneal injection of L-arginine to replicate the SAP model,Huoxue qingjie decoction group was treated with Huoxue qingjie decoction,control group was not processed.The changes of serum HO-1 and pyrodyosis factor(NLPR3,interleukin-1β[IL-1β])and the genes and proteins of HO-1,NLPR3 and IL-1βin pancreatic tissues of each group were compared.Results There were significant differences of the activity of HO-1 among three groups of time and inter-group(P<0.05);and there were no significant differences of the activity of HO-1 among three groups of interaction.At 24,48 and 72 h after treatment,the activity of HO-1 in the SAP model group was higher than that in the control group,and the difference was statistically significant(P<0.05).At 24,48 and 72 h after treatment,the activity of HO-1 in the Huoxue qingjie decoction group was higher than that in the SAP model group and the control group,the SAP model group was higher than the control group,and the differences were statistically significant(P<0.05).There were significant differences of the levels of IL-1βand IL-18 among three groups in inter-group,time and interaction(P<0.05).At 24,48 and 72 h after treatment,the levels of IL-1βand IL-18 in serum and pancreatic tissues in the SAP model group were higher than those in the control group,and the Huoxue qingjie decoction group were higher than those in the control group(P<0.05).At 24,48 and 72 h after treatment,the serum and pancreatic tissues of HO-1,NLPR3 and IL-1βlevels in the SAP model group were higher than those in the control group(P<0.05);the leve
