摘 要:目的探讨Ras同源基因家庭成员Ⅰ(ARHⅠ)、死亡相关蛋白激酶(DAPK)在皮肤鳞癌中的表达及意义。方法通过免疫组化法检测正常皮肤组织、Bowen病组织、皮肤鳞癌组织中ARHⅠ、DAPK阳性表达情况,通过实时荧光定量聚合酶链式反应检测正常皮肤组织、Bowen病组织、皮肤鳞癌组织中ARHⅠmRNA、DAPK mRNA相对表达量。分析ARHⅠmRNA、DAPK mRNA相对表达量与皮肤鳞癌严重程度的关系。结果正常皮肤组织、Bowen病组织、皮肤鳞癌组织中ARHⅠ、DAPK阳性表达细胞依次减少,Bowen病组织、皮肤鳞癌组织中的ARHⅠ、DAPK阳性表达率低于正常皮肤组织(P<0.05)。Bowen病组织、皮肤鳞癌组织中的ARHⅠmRNA、DAPK mRNA相对表达量少于正常皮肤组织(P<0.05)。随着皮肤鳞癌的加重,ARHⅠmRNA、DAPK mRNA相对表达量降低,其与皮肤鳞癌严重程度均呈负相关(r=-0.940、-0.932,P<0.01)。结论ARHⅠ、DAPK在皮肤鳞癌组织中下调或缺失,可能与皮肤鳞癌的发生、发展有重要关系。Objective To investigate the expression and significance of aplysia Ras homologⅠ(ARHⅠ)and death-associated protein kinase(DAPK)in skin squamous cell carcinoma.Methods The positive expressions of ARHⅠand DAPK in normal skin tissues,Bowen disease tissues and skin squamous cell carcinoma tissues were detected by immunohistochemistry;the relative expressions of ARHⅠmRNA and DAPK mRNA in normal skin tissues,Bowen disease tissues and skin squamous cell carcinoma tissues were detected by real-time fluorescence quantitative polymerase chain reaction.The relationship between relative expressions of ARHⅠmRNA and DAPK mRNA and severity of skin squamous cell carcinoma were analyzed.Results The positive expression cells of ARHⅠand DAPK in normal skin tissues,Bowen disease tissues and skin squamous cell carcinoma tissues decreased in turn;the positive expression rates of ARHⅠand DAPK in Bowen disease tissues and skin squamous cell carcinoma tissues were lower than those in normal skin tissue(P<0.05);the relative expressions of ARHⅠmRNA and DAPK mRNA in Bowen disease tissues and skin squamous cell carcinoma tissues were lower than those in normal skin tissues(P<0.05).With the aggravation of skin squamous cell carcinoma,the relative expressions of ARHⅠmRNA and DAPK mRNA decreased,which were negatively correlated with severity of skin squamous cell carcinoma(r=-0.940,-0.932,P<0.01).Conclusion The down-regulation or deletion of ARHⅠand DAPK in skin squamous cell carcinoma,may be related to the occurrence and development of skin squamous cell carcinoma.
