摘 要:目的利用生物信息学方法探讨HPV在口咽癌发生发展中的作用及其机制。方法将甲基化微阵列数据及转录组数据结合患者临床信息联合分析,解析口咽癌中HPV诱导的甲基化驱动基因及其生物学功能。结果富集分析显示HPV^(+)口咽癌患者的免疫活动明显激活,抗肿瘤T细胞免疫功能尤为活跃,这可能为HPV^(+)口咽癌患者更好的临床预后提供了一个合理的解释;CCR7和CXCR3可能是具有重大预后价值的HPV相关甲基化驱动基因。结论本研究揭示了HPV诱导的新型抗肿瘤免疫机制,为HPV^(+)与HPV^(-)口咽癌患者的预后差异提供了可能的依据,并提出HPV相关甲基化驱动基因可能是预测口咽癌患者临床预后的有效指标。Objective To explore the role of human pa pillomavirus(HPV)in the development of oropharyngeal cancer,and its mechanisms.Methods In this study,we analyzed methylation microarray data and transcriptomic sequencing data in con-junction with clinical information to decipher the methylation-driven genes induced by HPV and their biological functions in oro-pharyngeal carcinoma.Results Enrichment analysis showed that immune activity in HPV^(+)patients was significantly activated,with particularly active anti-tumor T cell immune function,providing a reasona ble explanation for the better clinical prognosis of HPV^(+)oropharyngeal cancer patients.CCR7 and CXCR3 were identified as HPV^(-)related methylation-driven genes with signifi-cant prognostic value.Conclusion This study revealed a novel anti-tumor immune function induced by HPV,providing possible evidence for the differential clinical outcomes of HPV^(+)oropharyngeal carcinoma patients,and suggesting that HPV^(-)related methylation-driven genes may serve as effective indicators for predicting clinical prognosis in oropharyngeal carcinoma patients.
