甜菜红素抑制大肠癌细胞转移相关基因的作用

摘  要:目的研究植物提取物甜菜红素对大肠癌细胞系HCT-116的杀伤作用以及抑制其转移和迁移的能力。方法使用单四氮唑法测定甜菜红素对大肠癌细胞系HCT-116的细胞毒性作用。用实时荧光定量PCR用于检测代表细胞转移表型和功能的多个基因表达。进行了与转移相对应的迁移和侵袭能力检测,以评估功能变化。结果甜菜红素在300μmol/L的浓度下作用3 d并未发现任何细胞杀伤作用和毒性效果。然而用甜菜红素处理的大肠癌细胞显示出促进细胞转移的3个基因Vimentin MMP-1 COX-2的表达量下降,另外促进细胞转移相关的3个基因VEGF EGFR L1CAM的表达量无变化。蛋白质定量分析和免疫荧光定位检测与定量PCR的结果基本一致。随后的功能实验分析表明:甜菜红素刺激的大肠癌细胞系迁移和侵袭能力下降。结论甜菜红素可以抑制大肠癌转移,有一定的抗肿瘤功能。Objective To study the killing effect of plant extract betanin on HCT-116 cell line of colorectal cancer and its ability to inhibit metastasis and migration.Methods The cytotoxicity of betanin on human colorectal cancer cell line HCT-116 was determined by MTT method.Quantitative real-time PCR was used to detect the expression of multiple genes representing cell metastasis phenotype and function.In addition,migration and invasiveness corresponding to metastasis were performed to assess functional changes.Results With the concentration of 300μmol/L,there was no cytotoxic effect and toxic effect for three days.The colorectal cancer cells treated with betanin showed a decrease in the expression of Vimentin MMP-1 COX-2,the three genes that promote cell metastasis.In addition,the expression of VEGF EGFR L1CAM,the three genes that promote cell migration and metastasis,remained unchanged.The results of protein quantitative analysis and immunofluorescence localization were basically consistent with those of quantitative PCR.The subsequent functional analysis showed that the migration and invasion of colorectal cancer cell lines stimulated by betanin decreased.Conclusion A certain concentration of betanin can inhibit the metastasis of colorectal cancer and has a certain anti-tumor function.
