摘  要:目的 观察塞来昔布对肝星状细胞增殖及胶原分泌的影响,探讨塞来昔布抗肝纤维化的作用机制。方法 将体外培养的肝星状细胞分为正常组、模型组和治疗组,模型组细胞给予PDGF刺激生长,治疗组细胞给予PDGF刺激后加入塞来昔布治疗,采用MTT法检测肝星状细胞的增殖,放射免疫法检测Ⅰ、Ⅲ型胶原含量。Western Blot法检测CyclinD1蛋白表达。结果 PDGF能够显著刺激肝星状细胞增殖和分泌Ⅰ、Ⅲ型胶原蛋白,塞来昔布不仅能够抑制经PDGF刺激的肝星状细胞的增殖,而且能够显著抑制肝星状细胞分泌Ⅰ、Ⅲ型胶原蛋白。塞来昔布抑制肝星状细胞CyclinD1蛋白表达。结论 塞来昔布可能通过下调CyclinD1蛋白表达抑制肝星状细胞的增殖和分泌胶原发挥抗肝纤维化的作用。Objective To discuss the role of celecoxi on the anti-fibrosis through observation of the effect of celecoxi on the proliferation and collagen secretion of hepatic stellate cells.Method Hepatic stellate cells were cultured in vitro and divided into normal group,model group and treatment group.Model group cells were stimulated with PDGF,treatment group cells were stimulated with PDGF and then treated with celecoxib.MTT was used to detect the proliferation of HSC,radioimmunoassay was used to analyse the secretion of collagen I and Ⅲ.Western Blot wasused to detect the expression of CyclinD1.Result PDGF significantly stimulated the growth of HSC and secretion of collagen Ⅰ and Ⅲ.Celecoxib not only dramatically inhibited the growth of HSC stimulated by PDGF,but also reduced the secretion of collagen Ⅰ and Ⅲ.Celecoxib reduced the expression of CyclinD1 in hepatic stellate cell.Conclusion Celecoxib maybe exert anti-fibrosis effect through down-regulation of CyclinD1 expression and inhibition of HSC growth and collagen secretion.
