摘 要:二酰甘油酰基转移酶(DGAT)在植物油脂合成过程中发挥重要作用。本研究利用实时荧光定量PCR检测方法,以低油低油酸品种花育17号、高油高油酸品种花育910和高油低油酸品种花育918为试材,分析了不同含油量花生品种不同发育时期种子中AhDGAT1-1、AhDGAT1-2和AhDGAT3-3基因的表达情况。结果表明:AhDGAT1-1基因表达峰值出现在低油低油酸品种和高油高油酸品种的荚果发育初期(下针后30天),而在高油低油酸品种中则出现在荚果发育中期(下针后40天);AhDGAT1-2基因的表达峰值出现在低油低油酸品种的荚果发育初期、高油高油酸品种和高油低油酸品种的籽仁油脂积累快速期;AhDGAT3-3基因在3个花生品种中的表达峰值均出现在荚果发育的中后期。推测AhDGAT1-1、AhDGAT1-2和AhDGAT3-3基因表达量的增加有利于花生油脂的合成与积累,但其起作用的时期存在种质差异。本研究结果可为花生高油、高油酸品种的选育提供理论依据。Diacylglycerol acyltransferase(DGAT)is a key enzyme in biosynthesis pathway of vegetable oil in the plant.Using the peanut varieties of Huayu 17 with low oil and low oleic acid,Huayu 910 with high oil and high oleic acid and Huayu 918 with high oil and low oleic acid as materials,the expression of Ah-DGAT1-1,AhDGAT1-2 and AhDGAT3-3 genes in peanut seeds at different developmental stages was analyzed by the quantitative real-time PCR.The results showed that the peak expression of AhDGAT1-1 gene appeared at the early pod developing stage(at 30 days after fruit-needle growth)in Huayu 17 and Huayu 910,while that in Huayu 918 appeared in the middle pod developing period(at 40 days after fruit-needle growth).The peak expression of AhDGAT1-2 gene appeared at the initial stage of pod developing in Huayu 17 and at the rapid stage of kernel oil accumulation in Huayu 910 and Huayu 918.The peak expression of AhDGAT3-3 gene the three varieties appeared at the middle and late stages of pod developing.It indicated that the increase of AhDGAT1-1,AhDGAT1-2 and AhDGAT3-3 expression level was beneficial to the synthesis and accumulation of lipids in peanut kernels,but their acting time was different among different varieties.These results could pro-vide theoretical bases for breeding peanut varieties with high oil and oleic acid contents.
