摘 要:试验旨在探究苦杏仁油对高脂血症模型小鼠体质量、血脂指标的影响。通过饲喂高脂饲料建立高血脂模型小鼠,经油脂灌胃干预,比较灌胃前后油脂剂量组与高脂模型组小鼠体质量、肝重、血清血脂水平、肝脏组织切片等指标。并采用离体的方法,模拟人体胃和肠道环境,研究油脂对胆酸盐的结合能力,来评价油脂降血脂的功能。结果表明,在1 mg/ml的浓度下,油脂对牛黄胆酸盐和甘氨胆酸盐的结合率分别达到了44.04%和53.31%,说明油脂具有较好的降血脂作用。结果显示,不同剂量的油脂对高血脂均有一定的改善,与高脂模型组相比,经油脂中剂量组灌胃后小鼠体重显著增长,中剂量油脂组处理显著降低总胆固醇(TC)至2.80 mmol/L、显著降低甘油三酯(TG)至1.59 mmol/L、显著降低低密度脂蛋白胆固醇(LDL-C)至2.59 mmol/L,极显著提升高密度脂蛋白胆固醇(HDL-C)至0.86 mmol/L。各剂量的油脂均能显著增加小鼠血清中超氧化物歧化酶的活性,显著降低血清中丙二醛的含量,缓解高脂饮食带来的肝脏损伤。研究表明,苦杏仁油能够调节由高脂饮食引起的肝细胞损伤和血脂升高。The effect of bitter almond oil on reducing blood lipid was studied in vivo and in vitro.In vitro method was used to simulate the environment of human stomach and intestine,study the binding ability of oil to cholate,and evaluate the function of oil in reducing blood lipid.The hyperlipidemia model was established by feeding high-fat diet.Through the intervention of oil gavage,the body mass,the contents of serum total cholesterol(TC),total triglyceride(TG),low-density total cholesterol(LDL-C),high-density total cholesterol(HDL-C),liver tissue sections and other indexes of the oil dose group and the high-fat model group were compared before and after gavage.The results showed that at the concentration of 1mg/ml,the binding rates of oil to bovine cholic acid salt and glycocholic acid salt could reach 44.04%and 53.31%respectively,indicating that oil has a good effect of reducing blood lipid in vitro.The experiment of reducing blood lipid in vivo showed that compared with the high-fat model group,oil significantly reduced the weight growth of high-fat mice.The treatment of medium dose oil group significantly reduced TC to 2.80 mmol/L,TG to 1.59 mmol/L,LDL-C to 2.59 mmol/L,and HDL-C to 0.86 mmol/L.All doses of oil could significantly increase the activity of superoxide dismutase in mouse serum,significantly reduced the content of malondialdehyde in serum,and alleviated the liver injury caused by high-fat diet.Both in vivo and in vitro results showed that bitter almond oil could reduce hepatocyte injury and elevated blood lipid caused by high-fat diet.
