摘 要:目的探讨补脾养心方对心脾两虚证失眠小鼠补益心脾和改善睡眠的作用。方法采用力竭游泳与灌服盐酸普萘洛尔溶液相结合模式复制病—证结合的心脾两虚证失眠小鼠模型,补脾养心方低(1.5 g/kg)、中(3 g/kg)、高(6 g/kg)剂量和阳性对照人参归脾丸(5 g/kg)每天灌胃,连续给药30天后检测各组小鼠体质量和摄食量、血清中谷丙转氨酶(alanine aminotransferase,ALT)、碱性磷酸酶(alkaline phosphatase,ALP)、谷草转氨酶(aspartate aminotransferase,AST)、乳酸脱氢酶(lactate dehydrogenase,LDH)和肌酸激酶(creatine kinase,CK)含量及胸腺、脾脏指数;苏木素—伊红染色法分析心、肝、脾、肌肉组织病理变化;比较力竭游泳时间和悬尾不动时间,观察小鼠睡眠时间和睡眠潜伏期,分析小鼠的睡眠情况。结果与空白对照组比较,模型组小鼠体质量和摄食量明显减少,力竭游泳时间明显缩短且其悬尾不动时间延长,脾脏和胸腺指数降低,睡眠时间明显缩短(P<0.05);与模型组相比,阳性对照组、补脾养心方各剂量组小鼠摄食量和体质量增加,力竭游泳时间延长,悬尾不动时间缩短,睡眠时间延长、睡眠潜伏期缩短,发生了阈下剂量的睡眠行为,各组小鼠均无产生直接睡眠(P>0.05)。与空白对照组比较,模型组小鼠ALT、AST、ALP含量明显升高;与模型组相比,阳性对照组、补脾养心方各剂量组小鼠胸腺指数和脾脏指数升高,血清中ALT、AST、ALP及LDH和CK含量均降低(P<0.05),心肝脾肌肉无明显病理学改变。结论补脾养心方对心脾两虚小鼠具有补益心脾的中医功效和改善睡眠的作用。Objective To explore the effect of Bupiyangxin decoction on the heart and spleen deficiency in mice with insomnia and improving sleep.Methods A mouse model of cardio-spleen deficiency insomnia combined with exhaustive swimming and propranolol hydrochloride solution was used to replicate the disease-certificate combination of disease and spleen,and the doses of low(1.5 g/kg),medium(3 g/kg),high(6 g/kg)doses and positive control Renshen Guipi pills(5 g/kg)were used to gavage every day,and the weight and food intake of mice in each group were detected after 30 days of continuous administration,serum alanine aminotransferase(alanine aminotransferase,ALT),alkaline phosphatase(alkaline phosphatase,ALP),aspartate aminotransferase(aspartate aminotransferase,AST),lactate dehydrogenase(lactate dehydrogenase,LDH)and creatine kinase(creatine kinase,CK)content and thymus and spleen index;HE staining was used to analyze pathological changes in heart,liver,spleen and muscle tissue.The exhausted swimming time and the hanging tail immobility time were compared,the sleep time and sleep latency of mice were observed,and the sleep of mice was analyzed.Results Compared with the blank control group,the body mass and food intake of mice in the model group were significantly reduced,the exhausted swimming time was significantly shortened,the hanging immobility time was prolonged,the spleen and thymus indices were reduced,and the sleep time was significantly shortened(P<0.05);Compared with the model group,the mice in the positive control group and the spleen nourishing heart group increased their food intake and body weight,extended their exhausted swimming time,shortened the hanging tail immobility time,prolonged sleep time,shortened sleep latency,and underwent sleep behavior at subthreshold doses,and none of the mice in each group produced direct sleep(P>0.05).Compared with the blank control group,the contents of ALT,AST and ALP in the model group were significantly increased.Compared with the model group,the thymus index and spleen in
