摘  要:目的:探讨盐酸小檗碱与大豆分离蛋白缓释凝胶对2型糖尿病(T2DM)小鼠的降糖作用及其对肠道菌群的影响。方法:将80只C57BL/6J小鼠随机分成正常组与高脂高糖饲料组,采用高脂高糖饮食联合链脲佐菌素建立T2DM小鼠模型,将造模成功的小鼠随机分为模型组,片剂组(100 mg·kg^(-1)),缓释凝胶高、低剂量组(100 mg·kg^(-1)、50 mg·kg^(-1)),连续给药4周,检测小鼠体质量、空腹血糖(FBG)、空腹胰岛素(FINS)及糖化血红蛋白(HbA1c),利用16S rRNA高通量测序技术进行肠道菌群结构分析。结果:片剂组与缓释凝胶组体质量下降较模型组缓慢(P<0.05),片剂组与缓释凝胶高剂量组较模型组FBG下降、FINS升高(P<0.05);Alpha多样性中,缓释凝胶高剂量组chao1指数、shannon指数升高(P<0.05),Beta多样性中各组肠道菌群完全分离;在门水平上,片剂组与缓释凝胶高剂量组疣微菌门、拟杆菌门丰度升高(P<0.05),厚壁菌门及厚壁菌门与拟杆菌门比值降低(P<0.05);在属水平上,片剂组与缓释凝胶组阿克曼氏菌属、拟杆菌属丰度升高(P<0.05),大肠杆菌属丰度降低(P<0.05)。结论:盐酸小檗碱与大豆分离蛋白缓释凝胶改善了T2DM小鼠肠道菌群结构,以上调阿克曼氏菌属为主,改善胰岛素抵抗程度,发挥降糖作用。OBJECTIVE To investigate the effects of berberine hydrochloride and soy isolated protein sustained release gel on intestinal flora in type 2 diabetic mice.METHODS Eighty C57BL/6J mice were randomly divided into normal group and high fat and sugar diet group.The high fat and sugar diet combined with streptozotocin(STZ)was used to establish T2DM mouse model,and the successfully modeling mice were randomly divided into model group,tablet group(100 mg·kg^(-1)),high-and low-dose sustained release gel(100 mg·kg^(-1) and 50 mg·kg^(-1))groups,and administered for 4 consecutive weeks.The body weight,fasting blood glucose(FBG),fasting insulin(FINS)and glycosylated hemoglobin(HbA1c)were detected in mice,and the intestinal flora structure was analyzed by 16S rRNA high-throughput sequencing technology.RESULTS Compared with that of the model group,the body weight of the tablet group and the sustained release gel groups decreased slowly(P<0.05),the FBG of the tablet group and the high-dose sustained release gel group decreased and the FINS increased(P<0.05);in Alpha diversity,the Chao 1 index and Shannon index of the high-dose gel group were higher(P<0.05).In Beta diversity,the intestinal flora of each group were completely separated.At the level of phylum,the abundance of Verrucomicrobiota and Bacteroidota increased(P<0.05),Firmicutes and the ratio of Firmicutes to Bacteroidota decreased(P<0.05)in the tablet group and the high-dose gel group.At the genus level,the abundance of Akkermansia and Bacteroides increased(P<0.05),and the abundance of Colidextribacter decreased(P<0.05)in tablet group and the sustained release gel groups.CONCLUSION Berberine hydrochloride and soy isolated protein sustained release gel can improve the structure of intestinal flora in T2DM mice,which mainly up-regulates Akkermansia,improves insulin resistance and exerts a hypoglycemic effect.
