摘  要:目的 研究分化型胚胎软骨发育基因2(DEC2)和缺氧诱导因子-2α(HIF-2α)在胃癌中的表达以及与临床病理参数之间的关系。方法 将胃癌细胞株MKN45和HGC27分为实验组(缺氧72h组)和对照组(常氧72 h组),检测各组细胞DEC2和HIF-2α mRNA及蛋白的表达;并在这两种胃癌细胞株中转染DEC2真核表达质粒,检测DEC2和HIF-2α蛋白水平的表达。另外,通过免疫组织化学染色的方法检测60例胃癌组织及相应的癌旁胃黏膜组织中DEC2和HIF-2α的表达,并分析其相关性。结果 缺氧处理后DEC2的mRNA及蛋白表达明显低于对照组,而HIF-2α的表达高于对照组。过表达DEC2后,HIF-2α的表达降低。DEC2在胃癌组织中的表达显著低于癌旁胃黏膜组织(χ^(2)=20.512,P<0.001),HIF-2α的表达则高于癌旁胃黏膜组织(χ^(2)=11.172,P=0.001)。在胃癌组织中DEC2、HIF-2α的表达水平与肿瘤的浸润深度、淋巴结转移及临床分期显著相关。DEC2与HIF-2α的表达在胃癌组织中呈负相关(P<0.001)。结论 DEC2在胃癌细胞及组织中低表达,HIF-2α在胃癌细胞及组织中高表达,两者呈负相关,且与肿瘤发展转移联系密切。Objective To investigate the expressions of differentiated embryonic chondrocyte gene 2(DEC2) and hypoxia inducible factor-2α(HIF-2α) and their relationship with the clinicopathological parameters of gastric cancer.Methods The gastric cancer cell lines MKN45 and HGC27 were divided into the experimental groups(hypoxia for 72 hours) and the control group(normoxic for 72 hours).The mRNA and protein expressions of DEC2 and HIF-2α were detected.The overexpression plasmid DEC2 was transfected into the cell lines to detect the protein expressions of DEC2 and HIF-2α.The expressions of DEC2 and HIF-2α in 60 cases of gastric cancer tissues and corresponding adjacent gastric mucosa tissues were detected with immunohistochemical staining,and their correlation was analyzed.Results After hypoxia treatment,the mRNA and protein expressions of DEC2 were significantly decreased in the experimental groups compared with the control groups,while the expressions of HIF-2α were increased.After overexpression of DEC2,the expression of HIF-2α decreased.The expression of DEC2 in gastric cancer tissues was significantly lower than that in adjacent gastric mucosa tissues(χ^(2)=20.512,P<0.001),while the expression of HIF-2α was higher(χ^(2)=11.172,P=0.001).The expressions of DEC2 and HIF-2α were significantly correlated with the depth of tumor invasion,lymph node metastasis and clinical stage,and the expressions of DEC2 and HIF-2α were negatively correlated with each other(P<0.001).Conclusion DEC2 is lowly expressed in gastric cancer cells and tissues,while HIF-2α is highly expressed.They are negatively correlated with other and closely related with the tumor development and metastasis.
