摘 要:目的研究花姜酮对人口腔鳞状细胞癌细胞上皮间质转化(EMT)的影响。方法采用不同浓度(0、5、10、20μmol/L)花姜酮处理人口腔鳞状细胞癌HSC-3细胞,分别在培养24、48、72 h时,应用实时定量荧光PCR检测EMT标志物波形蛋白(Vimentin)和E-钙黏蛋白(E-cadherin)mRNA表达水平,并于培养48 h时采用Western blot法检测Vimentin和E-cadherin蛋白表达水平;采用MTT检测细胞增殖能力、流式细胞仪测定细胞凋亡能力、Transwell小室测定细胞侵袭能力。结果花姜酮处理后,HSC-3细胞E-cadherin mRNA表达水平显著提高(P<0.05);HSC-3细胞Vimentin mRNA表达受到明显抑制,且具有浓度依赖性(P<0.05)。花姜酮处理48 h时,随着花姜酮浓度的增加,HSC-3细胞中Vimentin蛋白表达水平逐渐降低,E-cadherin蛋白表达水平逐渐升高(P<0.05)。随着花姜酮作用时间的延长及花姜酮浓度的增加,HSC-3细胞的增殖率逐渐降低(P<0.05)。花姜酮处理后,HSC-3细胞凋亡率明显升高,且随着花姜酮浓度的增加凋亡率显著升高(P<0.05);花姜酮处理后,HSC-3细胞侵袭数显著减少,随着花姜酮浓度的增加细胞侵袭数逐渐减少(P<0.05)。结论花姜酮可以通过抑制人口腔鳞状细胞癌细胞的增殖和侵袭,促进其凋亡,进而抑制EMT进程。Objective To study the effects of Gingerone on epithelial mesenchymal transformation(EMT)of human oral squamous cell carcinoma(OSCC)cells.Methods Human OSCC HSC-3 cells were treated with different concentrations of Gingerone(0,5,10,20μmol/L),and cultured for 24,48,72 h,respectively.The mRNA expression levels of EMT markers Vimentin and E-cadherin were detected by real-time quantitative PCR,and the protein expression levels of Vimentin and E-cadherin were detected by Western blot at 48 h after culture.Cell proliferation capacity was determined by MTT,cell apoptosis capacity was determined by flow cytometry,and cell invasion capacity was determined by Transwell chamber.Results The mRNA expression level of E-cadherin in HSC-3 cells was significantly increased after treatment with Gingerone(P<0.05).Vimentin mRNA expression in HSC-3 cells was significantly inhibited in a concentration-dependent manner(P<0.05).At 48 h after treatment with Gingerone,Vimentin protein expression level in HSC-3 cells was gradually decreased and E-cadherin protein expression level was gradually increased with the increase of Gingerone concentration(P<0.05).The proliferation rate of HSC-3 cells was decreased gradually with the prolongation of treatment time and the increase of Gingerone concentration(P<0.05).After treatment with Gingerone,the apoptosis rate of HSC-3 cells was notably increased,and the apoptosis rate was significantly increased with the increase of Gingerone concentration(P<0.05).The invasion number of HSC-3 cells was significantly decreased after treatment with Gingerone,and gradually decreased with the increase of Gingerone concentration(P<0.05).Conclusion Gingerone can inhibit the proliferation and invasion of human OSCC cells,promote their apoptosis,and inhibit the EMT process.
