摘 要:【目的】探究厚壳贻贝活性肽(Mytilus coruscus peptides,MCP)对酒精诱导小鼠肝损伤的保护及肠道菌群的调节作用。【方法】将45只C57BL/6小鼠平均分成5组:正常组、模型组(无水乙醇EtOH)、阳性组(EtOH+50 mg/kg水飞蓟素)、MCP低剂量组(EtOH+200 mg/kg MCP)和高剂量组(EtOH+400 mg/kg MCP)。Lieber DeCarli液体饲料喂养C57BL/6小鼠3周建立酒精性肝损伤模型,检测小鼠血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性及肝脏中抗氧化酶活性和抗炎因子水平,通过H&E染色和油红O染色观察肝脏组织形态变化和脂肪堆积情况,采用16s rRNA测序检测小鼠肠道菌群多样性变化。【结果】MCP显著提高肝脏中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)活性,显著降低促炎因子肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、白细胞介素1β(IL-1β)含量,同时降低了肝脏中甘油三酯(TG)和丙二醛(MDA)的含量(P<0.05);减少肝脏脂肪空泡,增加结肠杯状细胞,恢复了肝细胞和结肠细胞正常结构。改善了由于酒精引起的肠道中操作分类单元(OTU)总数增加的情况,并且降低了革兰阴性菌的比例。【结论】MCP对小鼠酒精性肝损伤具有明显的保护作用,同时能调节小鼠肠道菌群的组成。【Objective】To explore the protective effect of Mytilus coruscus peptides(MCP)on alcohol-induced liver injury and their regulation of gut microbiota in mice.【Method】Forty-five C57BL/6 mice were divided into five groups:normal group,model group(Anhydrous ethanol,EtOH),positive group(EtOH+50 mg/kg silymarin),MCP low-dose group(EtOH+200 mg/kg MCP)and high-dose group(EtOH+400 mg/kg MCP).C57BL/6 mice were fed with Lieber-DeCarli liquid diet for three weeks to establish alcoholic liver injury model.The activities of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum,antioxidant enzymes,and anti-inflammatory factors in liver were measured.H&E staining and oil red O staining were used to observe the changes of liver tissue morphology and fat accumulation,and the changes of gut microbiota diversity were detected.【Result】MCP significantly increased the activities of superoxide dismutase(SOD)and catalase(CAT)in the liver.It also significantly decreased the contents of pro-inflammatory cytokines tumor necrosis factorα(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β),as well as the contents of triglyceride(TG)and malondialdehyde(MDA)in the liver(P<0.05).MCP reduced the liver fat vacuoles,increased the colon goblet cells,and restored the normal structure of liver and colon cells.MCP also reduced the alcohol-induced operational taxonomic unit(OTUs)in the gut and reduced the proportion of gram-negative bacteria.【Conclusion】MCP has a protective effect against alcohol-induced liver injury in mice and can regulate the composition of gut microbiota.
