摘 要:采集六盘水煤矸山优势植物两头毛根际土壤进行培养,从含50 mg/L Cd^(2+)浓度培养基中分离出8种耐镉菌株。经不同Cd^(2+)浓度梯度和生长情况筛选得到耐镉能力较强和生长速度较快的真菌2株(P-4和P-5),其耐镉浓度分别高达900 mg/L和1000 mg/L。经形态学观察和ITS序列鉴定分别为被孢霉属(Mortierella)和拟盘多毛孢属菌(Pestalotiopsis)菌株。这两种耐镉菌株分别在不同pH(5、6、7、8、9)和温度(20℃、25℃、30℃)下培养生长,测定其菌落直径。结果表明,菌株P-4生长的最适pH为5,其菌落直径随温度升高而增大;菌株P-5的最适pH为6,最适温度为25℃。这些研究结果为以后镉污染土壤微生物修复提供了宝贵的微生物资源。Eight cadmium resistant fungi were isolated from the rhizosphere soil of Incarvillea arguta,a dominant plant grow⁃ing on the coal gangue pile in Liupanshui based on a medium containing 50 mg/L Cd^(2+).Two fungi(P-4 and P-5)were screened through different Cd^(2+)concentration gradients and growth conditions,with cadmium tolerance concentrations as high as 900 and 1000 mg/L,respectively.They were identified as Mortierella sp.and Pestalotiopsis sp.based on ITS segment.The results of colony diameter measurements of the two cadmium resistant strains grown in different pH(5,6,7,8,9)media and temperatures(20℃,25℃,30℃)showed that the optimal pH for the growth of strain P-4 was 5,and its colony diameter increased with the increase of temperature;The optimal pH of strain P-5 was 6,and the optimal temperature was 25℃.These research results provide valuable microbial resources for microbial remediation of cadmium contaminated soil in the future.
