摘 要:环境条件是影响生物表面活性剂强化甲烷氧化菌氧化甲烷效果的关键因素。选用鼠李糖脂为代表性生物表面活性剂,以甲烷氧化菌甲基弯菌(Methylosinus trichosporium)OB3b为供试菌株,以温度、pH及O_(2)、甲烷体积比(O_(2)/甲烷)为试验因素,采用单因素试验和响应面优化试验相结合的研究方法,开展了不同环境因素对鼠李糖脂提高甲基弯菌OB3b生长量(以600 nm处吸光度(OD_(600))表征)和甲烷氧化效率的影响研究。单因素试验结果表明:当温度为20~40℃、pH为5.0~9.0、O_(2)/甲烷为0~2.0,添加2倍临界胶束浓度(简写为2 CMC)鼠李糖脂均能够显著提高甲基弯菌OB3b培养过程的OD_(600)和甲烷氧化效率(p<0.05)。响应面优化试验结果表明:甲基弯菌OB3b的最佳培养条件为温度30℃、菌液pH=7.0、O_(2)/甲烷1.0,在该条件下添加2 CMC鼠李糖脂,甲基弯菌OB3b培养5 d的甲烷氧化效率能达95%左右。研究结果可为生物表面活性剂鼠李糖脂在生物除甲烷领域的应用提供理论基础和数据支持。Environmental conditions are the key factors affecting the promotion effect of methane oxidation by biosurfactants.This study selected rhamnolipid as the representative biosurfactant,and investigated the effects of different environmental factors on the growth(measured as OD_(600))and methane oxidation efficiency of Methylosinus trichosporium OB3b with the application of rhamnolipid by single factor and response surface optimization tests,in which temperature,pH and O_(2)/methane(volume ratio)in the culture environment were selected as the experimental factors.The single factor tests indicated that OD_(600) and methane oxidation efficiency of Methylosinus trichosporium OB3b were significantly improved(p<0.05)with the addition of rhamnolipid at a concentration of 2 CMC when the temperature of 20-40℃,pH of 5.0-9.0 and O_(2)/methane of 0-2.0.The response surface optimization test showed that the optimal culture conditions of Methylosinus trichosporium OB3b with 2 CMC rhamnolipid application were as follows:temperature of 30℃,pH of bacteria solution 7.0,and O_(2)/methane of 1.0.Under the above condition,the methane oxidation efficiency of Methylosinus trichosporium OB3b could reach about 95% after 5 d cultivation.The results can provide a theoretical basis and data support for the application of biosurfactant rhamnolipid in biological methane removal technologies.
