摘 要:目的探讨葡萄籽原花青素提取物(grape seed proanthocyanidins extract,GSPE)能否通过Nrf2信号通路拮抗砷所致的小鼠肝脏氧化损伤,为GSPE对抗砷的氧化损伤作用提供理论依据。方法以昆明种小鼠为研究对象,根据As 2O 3(0、4 mg/kg)和GSPE(0、400 mg/kg)的剂量,采用2×2析因设计方差分析,按照体质量分层,随机分为对照组、As 2O 3组、GSPE组和As 2O 3组+GSPE组,使用析因设计分析As 2O 3和GSPE的交互作用和As 2O 3、GSPE的单独效应。采用试剂盒标准的测量方法测量MDA、GSH、ROS、GSH-Px、SOD、CAT、LPO、-SH和T-AOC表达;Western blotting检测Nrf2蛋白的表达;qRT-PCR检测Nrf2 mRNA的表达水平。结果As 2O 3组中,小鼠的体质量有所下降,肝脏重量和脏器系数增加;GSPE可以对抗这一结果;As 2O 3可以增加MDA、ROS、LPO、Nrf2蛋白和mRNA的表达,减少GSH、GSH-Px、SOD、CAT、-SH和T-AOC的表达;在经过GSPE拮抗As 2O 3的作用之后,同组相比,MDA、ROS、LPO、Nrf2蛋白和mRNA的表达减少,而GSH、GSH-Px、SOD、CAT、-SH和T-AOC的表达增加(P均<0.05)。结论GSPE可能通过Nrf2信号通路及相关氧化应激因子拮抗砷所致的小鼠肝脏氧化损伤。Objective To explore whether grape seed proanthocyanidins extract(GSPE)can antagonize arsenic induced liver oxidative damage in mice through Nrf2 signaling pathway,so as to provide a theoretical basis for GSPE to antagonize arsenic induced oxidative damage.Methods Kunming mice were taken as the research object.According to the doses of As 2O 3(0,4 mg/kg)and GSPE(0,400 mg/kg),2×2 factorial design analysis of variance was used.Stratified by body weight,they were randomly divided into control group,As 2O 3 group,GSPE group and As 2O 3+GSPE group.Factorial design was used to analyze the interaction between As 2O 3 and GSPE,and the separate effects of As 2O 3 and GSPE.The expressions of MDA,GSH,ROS,GSH-Px,SOD,CAT,LPO,-SH and T-AOC were measured by the standard measurement method of the kit;the expression of Nrf2 protein was detected by Western blotting;qRT-PCR was used to detect the expression level of Nrf2 mRNA.Results In As 2O 3 group,the weight of mice decreased,and the liver weight and organ coefficient increased;GSPE could resist this result.As 2O 3 could increase the expression of MDA,ROS,LPO,Nrf2 protein and Nrf2 mRNA,and reduce the expression of GSH,GSH-Px,SOD,CAT,-SH and T-AOC.After GSPE antagonized the effect of As 2O 3,compared with the same group,the expression of MDA,ROS,LPO,Nrf2 protein and Nrf2 mRNA decreased,while the expression of GSH,GSH-Px,SOD,CAT,-SH and T-AOC increased(P<0.05).Conclusion GSPE may antagonize arsenic induced liver oxidative damage in mice through Nrf2 signaling pathway and related oxidative stress factors.
