摘  要:目的观察共同性外斜视猫内直肌中脑源性神经营养因子(BDNF)蛋白及mRNA的表达变化,并探讨其与共同性外斜视发病的相关性。方法28只雌性猫随机分为A、B、C组及对照组,每组各7只。A、B和C组猫分别全天配戴15△棱镜2、4、6个月,对照组猫全天配戴平光镜6个月。麻醉各组猫后取眼内直肌组织,Masson染色后测算猫眼内直肌纤维横截面积及肌卫星细胞数,分别采用RT-qPCR法和WESTERN Blotting法检测各组猫眼内直肌BDNF mRNA和蛋白。结果与对照组比较,A、B、C组猫的眼内直肌肌纤维横截面积小、肌卫星细胞数少(P均<0.05)。A、B、C组及对照组猫的眼内直肌BDNF mRNA相对表达量分别为0.51±0.05、0.41±0.04、0.10±0.04、1.00±0.10,BDNF蛋白相对表达量分别为0.85±0.11、0.66±0.05、0.14±0.05、0.97±0.09,与对照组比较,A、B、C组猫的眼内直肌BDNF mRNA及蛋白相对表达量低(P均<0.05)。结论共同性外斜视猫的眼内直肌组织BDNF mRNA及蛋白表达低。BDNF mRNA及蛋白可能在共同性外斜视的发生发展中发挥重要作用。Objective To investigate the protein and mRNA expression levels of brain-derived neurotrophic factor(BDNF)in concomitant exotropia cats,and to discuss their association with the development of concomitant exotropia.Methods Twenty-eight female cats(3-4 weeks old,weight 280-325 g)were randomly divided into the experimental groups A,B,C,and control group,with 7 in each.The cats in the experimental groups A,B and C were fitted with 15△prisms all day for 2,4 and 6 months,respectively.The cats in the the control group were fitted with flat lenses all day for 6 months.Masson staining was used to measure the cross-sectional area of muscle fibers and the number of muscle satellite cells of medial rectus muscle.RT-qPCR and Western blotting were used to detect the mRNA and protein expression of BDNF.Results Compared with the control group,the cross-sectional area of muscle fibers and the number of muscle satellite cells in the experimental group were significantly reduced in the experimental groups A,B,and C.The rela⁃tive mRNA expression levels of BDNF in medial rectus muscle of the control group and experimental groups A,B,and C were 1.00±0.10,0.51±0.05,0.41±0.04,and 0.10±0.04,respectively,and the relative protein expression levels of BDNF in medial rectus muscle of the control group and experimental groups A,B,and C were 0.97±0.09,0.85±0.11,0.66±0.05,and 0.14±0.05,respectively(all P<0.05).Compared with the control group,the relative expression lev⁃els of BDNF mRNA and protein in medial rectus muscle of experimental groups A,B and C were lower(all P<0.05).Conclusions The expression levels of BDNF mRNA and protein in the medial rectus muscle of cats with concomitant exo⁃tropia were low.BDNF mRNA and protein may play an important role in the development and progression of concomitant exotropia.
