摘　　要：目的探讨醋酸落叶松酯在体内外对膀胱癌的抗瘤作用。方法采用CCK-8实验、平板克隆实验、Western blotting及流式细胞术检测醋酸落叶松酯对体外培养的膀胱癌细胞生物学行为的作用。将醋酸落叶松酯注入膀胱癌原位移植瘤动物模型中,观察其在体内对膀胱癌的作用。钙成像技术观察醋酸落叶松酯对钙离子通道的作用。结果CCK-8实验、平板克隆实验证明,醋酸落叶松酯在体外能抑制膀胱癌T24和J82细胞的增殖,Western blotting实验及流式细胞术证明,醋酸落叶松酯可诱导T24和J82细胞体外凋亡。钙成像技术检测到醋酸落叶松酯对TRPC6的抑制作用。动物实验表明,醋酸落叶松酯在体内可抑制T24细胞增殖,从而抑制肿瘤生长。结论醋酸落叶松酯可在体内外抑制膀胱癌T24和J82细胞的增殖,并诱导其凋亡。Objective To investigate the anti-tumor effect of larixyl acetate(LA)on bladder cancer in vitro and in vivo.Methods CCK-8 assay,plate cloning assay,western blotting assay,and flow cytometry were used to detect the effect of larixyl acetate on the biological behavior of bladder cancer cells cultured in vitro.Observe the impact of larixyl acetate on bladder cancer in vivo by injecting it into the animal model of bladder carcinoma orthotopic transplantation tumor.Ca^(2+)imaging technique was applied to observe the effect of larixyl acetate on Ca^(2+)channels.Results The results of CCK-8 and plate cloning assay showed that larixyl acetate could inhibit the proliferation of bladder cancer T24 and J82 cells in vitro.The Western blotting and flow cytometry results showed that larixyl acetate could induce the apoptosis of T24 and J82 cells in vitro.Ca^(2+)imaging analysis showed the inhibitory effect of larixyl acetate on TRPC6.In vivo experiments showed that larixyl acetate inhibited the proliferation of T24 cells,and thus inhibited tumor growth.Conclusion Larixyl acetate can inhibit the proliferation of human bladder cancer T24 and J82 cells in vitro and in vivo,and induce the apoptosis of T24 and J82 cells in vitro.
