摘 要:目的探讨环状RNA circ_0000264在乳腺癌细胞增殖、迁移和侵袭中的功能及作用机制。方法采用qRT-PCR检测乳腺癌组织和细胞系中circ_0000264的表达;利用si-circ_0000264构建敲低circ_000026的细胞模型;分别采用CCK-8、BrdU和Transwell实验检测细胞的增殖、迁移和侵袭;采用双荧光素酶报告基因实验验证circ_0000264与miR-622的靶向调控关系。结果circ_0000264在乳腺癌组织和细胞中均呈高表达(P<0.05)。与对照组相比,敲低circ_0000264可抑制乳腺癌细胞的增殖、迁移和侵袭(P<0.05)。circ_0000264对miR-622具有吸附作用。下调miR-622可部分逆转敲低circ_0000264对乳腺癌细胞增殖和转移的抑制作用。结论circ_0000264在乳腺癌进展过程中发挥促进作用;circ_0000264通过负向调控miR-622促进乳腺癌细胞的增殖、迁移和侵袭。Objective To investigate the function and mechanism of circular RNA circ_0000264 in the proliferation,migration and invasion of breast cancer cells.Methods The qRT-PCR was used to detect the expression of circ_0000264 in breast cancer tissues and cell lines.The si-circ_0000264 was adopted to construct a cell model with low expression of circ_0000264.The cell proliferation,migration and invasion abilities were detected by CCK-8,BrdU and Transwell assay,respectively.The targeted regulation relationship between circ_0000264 and miR-622 was verified by dual-luciferase reporter gene assay.Results The circ_0000264 was highly expressed in both breast cancer tissues and cells.Compared with the control group,knocking down circ_0000264 could inhibit the proliferation,migration and invasion of breast cancer cells.The potential mechanism confirmed that circ_0000264 adsorbed miR-622.In addition,down-regulation of miR-622 could partially weaken the inhibitory effect of knocking down circ_0000264 on the proliferation and metastasis of breast cancer cells.Conclusion The circ_0000264 played an important role in the progression of breast cancer.It promoted the proliferation,migration and invasion of breast cancer cells by negatively regulating miR-622.
