摘　　要：目的探讨胞质分裂蛋白调节因子1(PRC1)对人肝内胆管细胞癌(ICC)中表达及作用,探究受PRC1影响的ICC发病机制。方法首先通过免疫组化验证PRC1在ICC中的表达情况。进而敲低ICC中PRC1的表达,通过CCK-8、Transwell、细胞凋亡检测等方法,测定PRC1在ICC中的作用,并通过Westernblot测定相关蛋白表达,探讨其可能的机制。结果PRC1在人ICC组织中表达升高。敲低PRC1后抑制ICC细胞的增殖、迁移和侵袭,促进ICC细胞的凋亡。结论PRC1在ICC中表达升高,其可能通过抑制ICC细胞的凋亡进而影响肿瘤细胞的活性。Objective To investigate the expression and function of PRC1 in intrahepatic cholangiocarcinoma(ICC)and to explore the pathogenesis of ICC.Methods The expression of PRC1 in ICC was detected by immunohistochemistry.Then the expression of PRC1 in ICC was knocked down,and the role of PRC1 in ICC was detected by CCK-8,Transwell and apoptosis detection methods.The expression of related proteins was detected by Westernblot to explore its possible mechanism.Results PRC1 expression was increased in ICC tissues.After PRC1 knockdown,the proliferation,migration and invasion of ICC cells were inhibited,and the apoptosis of ICC cells was promoted.Conclusion The expression of PRC1 is increased in ICC,which may affect the activity of ICC cells by inhibiting the apoptosis of ICC cells.
