基于线粒体COI及16S rRNA基因序列的笠贝科系统进化研究

摘  要:采集了中国东南沿海地区笠贝科3属7种笠贝样品:陆川小笠贝、北戴河小笠贝Nipponacmea sp.、黑新笠贝、鸡爪拟帽贝、琉球拟帽贝,通过PCR扩增笠贝的COI及16S rRNA基因片段并测序,另外从GenBank中下载了笠贝科11种笠贝的COI和16S rRNA基因,计算了所有18种笠贝的碱基含量和遗传距离,并分别构建了基于COI和16S rRNA系统进化树。结果表明:COI基因片段长度为550 bp,16S rRNA基因片段长度为449 bp,T碱基含量最高,C碱基含量最少,COI基因的遗传距离为0.004~0.470,16S rRNA的遗传距离为0.013~0.575,系统发育分析表明,3个属的进化关系为Nipponacmea与Lottia互为姐妹群,其次为Patelloida。每个属都形成独立分支,参与分析的其他属的进化地位也与早期研究相接近,但本研究中,COI和16S rRNA的系统发育树也呈现出略微差异,如笠贝科的Yayoiacmea与Tectura聚集在一起形成了姐妹群,Patelloida内物种的位置也产生了小范围的变化。In this study,seven species of Lottiidae belonging to three genera were collected from the coastal areas of southeast China:Lottia luchuana,Lottia peitaihoensis,Nipponacmea sp.,Nipponacmea nigrans,Nipponacmea fuscovidis,Patelloida saccharinoides,and Patelloidar yukyuensis.The COI and 16S rRNA gene fragments of the samples were amplified and sequenced using PCR.Additionally,COI and 16S rRNA genes from 11 other Lottiidae species were downloaded from GenBank.The base content and genetic distance of all 18 Lottiidae species were calculated.The phylogenetic trees based on COI and 16S rRNA were constructed respectively.The results showed that the length of COI gene segment was 550 bp,the length of 16S rRNA gene segment was 449 bp,the content of T base was the highest,the content of C base was the lowest,the genetic distance of COI gene was 0.004-0.470,and the genetic distance of 16S rRNA was 0.013-0.575.Phylogenetic analysis showed that Nipponacmea and Lottia were sister groups,followed by Patellida.Each genus forms an independent branch,and the evolutionary status of other genera involved in the analysis is also close to that of earlier studies.However,in this study,the phylogenetic tree of COI and 16S rRNA also shows a slight difference.For example,the Yayoiacmea and Tectura genera of the Lottiidae gather together to form a sister group,and the position of species in the Patellida genus also changed slightly.
