姜黄素对人结肠癌肿瘤样干细胞中VEGF表达的作用

摘　　要：目的研究姜黄素对体外培养的人结肠癌细胞株HT-29、SW480中VEGF表达的影响,探讨姜黄素抑制肿瘤细胞生长机制,为姜黄素抗结肠癌治疗提供一定的实验依据。方法利用荧光显微镜观察姜黄素处理后细胞生长及增殖情况变化;利用实时定量PCR(Real-time PCR)检测姜黄素处理后HT-29、SW480细胞中VEGF的mRNA表达。利用蛋白免疫印迹(Western-blot)检测姜黄素处理后HT-29、SW480细胞中VEGF蛋白表达的变化。结果与空白对照组相比,姜黄素处理后的人结肠癌细胞数目明显减少,生长速度减慢;在基因和蛋白水平VEGF表达明显降低,差异有统计学意义(P<0.05)。结论姜黄素可抑制体外培养的人结肠癌细胞的生长及增殖,同时抑制VEGF基因及蛋白表达,为进一步明确姜黄素抑制结肠癌细胞生长和增殖及转移机制奠定基础。Objective To study Curcumin in culture of colon cancer cell lines HT-29,SW480 cells,the influence of vascular endothelial growth factor(VEGF)expression,for clinical treatment of colon cancer provide theoretical and experimental basis.Methods Cell growth and proliferation of Curcumin were observed by fluorescence microscope,and the mRNA expression of VEGF in HT-29 and SW 480 cells was measured by real-time PCR(Real-time PCR).The VEGF mRNA expression change in HT-29 and SW 480 cells after Curcumin treatment was determined by protein immunoblotting(Western-blot).Results In mRNA expression level,Curcumin processing HT-29 cell lines,compared with the control,Curcumin processing SW480 cell lines,VEGF expression level was lower,and there were statistical statistically differences(P<0.05).Conclusion Curcumin in vitro culture can inhibit the growth and proliferation of human colon cancer cell,and also inhibit the expression VEGF gene and the protein expression.laying a foundation for further clarifying the mechanism of Curcumin in inhibiting the growth,proliferation and metastasis of colon cancer cells.
